| Objective: To investigate the effect of Shenqi antimicrobial on the tumor of mice bearing tumor of MFC,and to explore its mechanism of tumor inhibition and the direction of apoptosis and invasion and metastasis of tumor cells.Methods: 50 BALB/c-nu mice are adaptive fed 7 days,and modelling by injecting MFC cell suspension in the right front underarm.It is indicated that the model is successful after the pathological nodules are diagnosed as tumor,which are derived from the randomly selected nodular mice,male and female 1 only.The rest of model mice were randomized into model group(M),cis-diamin-odichlor platinum group(DDP),low dosage of Chinese medicine group(TCML),high dosage of Chinese medicine group(TCMH),low dosage of combination group(TCML+DDP),high dosage of combination group(TCMH+DDP),with 8 animals in each group.Another 8 healthy mice are regarded as normal control group(N).The therapy of Chinese medicine lasted for 15 days,1 times a day,while chemotherapy were given at the first and eighth days.Complete the spleen and tumor tissue.Observe the normal and tumourconditions.Tumor and spleen were obtained from mice for tumor inhibition rate,q value,spleen index,pathological detection of tumor tissue,immunohistochemistry detection of tumor tissue TGF-?,MMP13,XIAP,PTEN protein and q RT-PCR detection of TGF-?,MMP13,XIAP,PTEN m RNA.Results:(1)After modeling,the tumor body gradually increased,the tumor surface from round to oval gradually changed into irregular shape,the activity was poor;The traditional Chinese medicine group and the combined group mice were generally better than the model group and cisplatin group.(2)Spleen index of TCML group is increased by 31.86%(P<0.01),while TCMH group is increased by 36.27% compared with M group(P<0.01).TCML+DDP group is increased by 19%(P<0.01),while TCMH+DDP group is increased by 39% compared with DDP group(P<0.01).(3)The tumor inhibiting rate of DDP group was 55.25%,while the Chinese medicine group inhibiting rate were 27.7%,30.58% respectively.At the same time,the inhibiting rates of combination group were 66.3%,67.78% respectively.TCML+DDP group was 24.7% higher than DDP group,and 53.39% higher than TCML group.TCMH+DDP group was 28% higher than DDP group,and 53.59% higher than TCMH.It was proved that both drugs used was better than single,the difference was statistically significant(P<0.01).Q value was calculated,q?0.942/0.983.(4)Pathological tissue showed that the number of cells in M group was much,and the atypia of tumor cells was obvious.The number of tumor cells in the treatment group decreased and the necrosis increased.(5)Compared with the M group,the expression of XIAP,TGF-?,MMP 13 m RNA and protein in DDP group,Chinese medicine group and combined group decreased,the expression of PTEN m RNA and protein increased,the difference was statistically significant(p < 0.01).(6)Compared with DDP group,TCML and TCMH group,the expression of XIAP,TGF-?,MMP 13 m RNA and protein decreased,the expression of PTEN m RNA and protein increased,the difference was statistically significant(p < 0.01).Conclusion: Shenqi Yiliu Decoction has a certain effect of anti-MFC gastric cancer transplant,and combined with cis-diamin-odichlor platinum has a synergistic effect.Its effect may be through Down the expression of TGF-?,MMP 13,XIAP,increase the expression of PTEN m RNA and protein,so as to inhibit tumor cell proliferation,inhibit tumor invasion and metastasis,and promote apoptosis of tumor cells. |
