Objective: To observe the effects of serum containing Fuzheng Yiliu Decoction on rat bone marrow mesenchymal stem cells(BMSCs) growth, apoptosis and telomerase activity and p53 expression in glioma(C6) environment.Methods: Using Transwell co culture of BMSCs and C6 cells, experiment set 4 groups:BMSCs control group(BMSCs + blank control serum), BMSCs contained serum group(BMSCs+ Fuzheng Yiliu Decoction containing serum), coculture control group(cocultured BMSCs cells and C6 cells + control serum), co culture contained serum group(cocultured BMSCs cells and C6 cells + Fuzheng Yiliu decoction containing serum), Fuzheng Yiliu Decoction containing 10%serum concentration was used for intervention.After incubated for 72 h, BMSCs were trypsinized and inoculated in 96 well plates, and then were cultured for 24 h, 48 h and 72 h, methyl thiazolyl tetrazolium method was used to assess the proliferation of BMSCs; the cell cycle and apoptosis rate of BMSCs were detected by flow cytometry, telomerase activity was detected by trap-PCR,and p53 protein expression was estimated by Western blot.Results: The cell morphology of the BMSCs at control group showed a long spindle shape,the colony-like distribution.BMSCs to the fixed nuclear cytoplasmic shrinkage after 72 h co culture,the cells became round, but not like glioma cell like morphology change. Fuzheng Yiliu Decoction serum intervention of co cultured BMSCs cells were fibroblast like morphology.Compared with the BMSCs control group, the proliferation rate of BMSCs in co-culture control group was increased with time, and also the proportion of cells in S phase, but the apoptosis rate was decreased, while the telomerase activity was increased(P≤ 0.05), the change of p53 protein expression level had no significant difference(P>0.05). Compared with the co-culture group, the proliferation rate of BMSCs in containing drug serum group was decreased in a time-dependent manner, the proportion of cells in S phase was reduced, cell apoptosis rate was increased, the telomerase activity was reduced, and p53 protein expression level was increased(P≤ 0.05).Conclusion:(1) After incubated for 72 h,Morphology changes of BMSCs were not fibroblast-like, but cells occured abnormal proliferation and the percentage of S phase wasincreased, telomerase activity was increased in C6 microenvironment.(2) The serum containing Fuzheng Yiliu Decoction had inhibitory effect on above changes.
