Objective: To observe the gastric bypass surgery(GBP)on the effects of spontaneous type 2 diabetic rats(GK rats)in plasma adiponectin(APN)and hepatic tissue insulin receptor-?(IR-?)and glucose transporter 2(GLUT-2)expression,and to explore the mechanism of improving insulin resistance.Methods: 30 male GK rats.Were randomly divided into three groups,respectively operation group,sham operation group and diet control group,rats in each group were10.At the same time,10 SD rats as blank control group.Surgical group received gastric bypass surgery way.The sham operation group underwent laparotomy,no surgery.Diet control group rats received surgery and drug treatment,only from the control diet and exercise.The blank control group without any treatment.Detection and 4 week APN in plasma were compared in rats before and after the operation.The rats were killed after 4 weeks of rat hepatic tissue were detected.WB experimental method was used to detect the expression of IR-? and GLUT-2 in rat hepatic tissue.Results: Operative group,after 4 weeks,the APN levels[(30.41±5.32)pmol/L] were higher than preoperative group(P < 0.05);As for GK operation group,the FPG level[(5.13±0.22)mmol/L] decreased significantly after the operation(P<0.05),and the HOMA-IR level[(2.16±0.18)mmol/L] decreased significantly too(P<0.01).The expressions of hepatic IR-? and GLUT-2 protein were significantly higher in GK operation group(P<0.01).Conclusion: 1.The level of APN in plasma of GK rats was significantly increased after GBP.2 The expressions of GLUT-2 and IR-? protein in hepatic tissue of GK rats after GBP was increased,and the symptoms of insulin resistance were relieved.
