Objective:To investigate the role of serum Adiponectin and the expression of Adiponectin receptor 2 in liver of impaired glucose tolerance rats.Methods:31 male Wistar rats were randomly distributed into two groups:control group and high fat-fed group.The model group were fed with high-fat feeding,to establish impaired glucose tolerance animal model.After high-fat feeding for 8 weeks,12 weeks,the blood samples were obtained to measure blood fat series,fast plasma glucose,Adiponectin,Fasting serum Insulin,assessed the degree of insulin resistance by using homeostasis model assessment of insulin resistance..The liver sample were used to analyse morphologically.Adiponectin and Adiponectin receptor 2 protein were respectively examined by ELISA and Western blotting.Results:1.Compared with the control group,the weight,VF/W,oral glucose test 2 hour plasma glucose,TG,TC and LDL were higher in high fat-fed group.In the meantime,HDL increasingly decreased.There was a significant difference between the two groups(p<0.05).After 12-week high-fat feeding,OGTT2hPG reached(8.29±0.27) mmol/ L,the rat model of IGT were produced,while its fasting blood glucose increased than control group(p<0.05) but not achieved the diagnostic criteria of impaired fasting blood glucose2.The plasma FINS and HOMA—IR concentrations of impaired glucose tolerance group increasingly elevated.Compared with the model group,the difference is significant in 8 week,12week.(å‡ä¸ºp<0.05).3.At 8,12 week,in high fat-fed group serum Adiponectin decreased than that in control group(p<0.05).4.The expression of Adiponectin receptor 2 in liver of impaired glucose tolerance rats is abundant.The adiponectin receptor 2 protein in liver decreased in high fat-fed group at 8,12 week,than that in control group(p<0.05)..5.The adiponectin receptor 2 protein in liver was correlated negatively with HOMA-IR,FINS,TC,TG,LDL,FPG and OGTT2hPG,positively with the serum Adiponectin and HDL.6.Liver sample were HE-stained.There was evidently fatty infiltration in liver of high fat-fed group group.It markedly differed from the control group.Conclusions: 1.Decrease adiponectin and Adiponectin receptor 2 correlated closely with the development of NASH.2.Adiponectin can be used as an early figure to predict non-alcoholic steatohepatitis. Adiponectin and Adiponectin receptor 2 can be used as molecule target to do intervention treatment in IGT and predict non-alcoholic steato-hepatitis. |
