ObjectivesAdiponectin is a protein secreted by adipocytes.It has been found that adiponectin has the effect of improving insulin resistance,lowering blood sugar,anti hypertension and alleviating inflammatory response.In this study,a rat model of type 2 diabetes mellitus was induced by intraperitoneal injection of low-dose streptozotocin(STZ)on the basis of SD rats' animal experiment.The expression of adiponectin and glucose transporter 4(GluT4)was observed under the intervention of hyperbaric oxygen(HBO)in the 2 type diabetic rats.To investigate the effect of hyperbaric oxygen(HBO)on the expression of adiponectin and GluT4 in type 2 diabetic rats in order to study the prevention and treatment of hyperbaric oxygen in type 2 diabetes and its complications Methods:10 healthy control subjects(group A,n=10)were randomly selected from a total of 40 male SD rats.They were fed with normal diet.The remaining 30 rats in the model group were fed with high-fat and high sugar diet for 4 weeks.Then the model group fasted for 12 hours,and small dose of streptozotocin(STZ)was injected intraperitoneally.Fasting blood glucose was measured after 72 hours of injection,21 rats of more than 16.7mmol/L were type 2 diabetes rats.The model of type 2 diabetic rats was randomly divided into diabetic control group(group B,n=11)and diabetic hyperbaric oxygen therapy group(group C,n=10).Group C was treated with hyperbaric oxygen for 60 minutes every day for 30 days under 0.1MP conditions.Group C was given hyperbaric oxygen(HBO)every day for 60 minutes under 0.1MP condition,which lasted for 30 days.After 30 days of hyperbaric oxygen intervention,3 rats from rats of each group were randomly selected for glucose tolerance test.Then the rats in each group were fasted for 12 hours,and they were weighed and anaesthetized.The abdominal aorta was taken for blood.After anticoagulation of heparin tube,centrifugation is reserved for serum reserve.The rats were sacrificed and the spleen was immediately cut and weighed,and the liver tissue of the soybean was cut.They were fixed with formalin solution and embedded in dehydrated paraffin.Next,they were routinely sliced for further preparation of HE staining for liver and spleen and immunohistochemical staining of liver.The serum adiponectin,insulin and GluT4 in each group were detected by ELISA kit.The serum high-density lipoprotein(HDL-C),low density lipoprotein(LDL-C),cholesterol(TC)and triglyceride(TG)were detected by automatic biochemical analyzer.Immunohistochemical staining was used to analyze the expression of adiponectin receptor 2(AdipoR2)and insulin receptor(INSR)in liver samples.Results:1.General situation: The rats in group A had good growth,normal feeding,quick response,smooth hair and glossy hair.However,the intake,diet and urine volume of B group increased significantly,accompanied by mental fatigue,slow action,and withered hair.The general situation of rats in group C was better than that in group B.2.The body weight of rats fed with high fat and high sugar feed for 4 weeks was significantly higher than that in the normal diet group(P<0.05),while the trend of FBG increased,but there was no statistical significance(P>0.05).3.After the experiment,Compared with group B,the weight of rats in group C was more than that of group B,and the weight of spleen was less than that of group B.At the same time,30 minutes after meal,blood glucose,60 minutes after meal,120 minutes of blood glucose,triglyceride and total cholesterol were decreased.But none of them were statistically significant.Fasting blood glucose and liver AdipoR2 decreased significantly(P<0.05),and the expression of high-density lipoprotein,insulin sensitivity,serum adiponectin and GluT4 increased significantly(P<0.05).4.Liver HE: Group B: Hepatocytes were enlarged,loose in cytoplasm,arranged in disorder,and steatosis in liver cells increased significantly.A large number of lipid droplets appear in the hepatocytes,and the nucleus is pushed to one side.There were punctate necrosis in the lobule of liver,infiltration of inflammatory cells and narrowing of the gap between the sinusoids.After 30 days of hyperbaric oxygen intervention,C group: hepatic steatosis improved significantly,and inflammatory cell infiltration decreased.The cell cord was arranged orderly,and the hepatic sinusoidal space was narrow.5.Spleen HE: Group B:the distribution of the spleen in the B group was irregular and the germinal center atrophied,and the C group was better than the B group.Conclusions:Hyperbaric oxygen therapy not only improves the content of adiponectin in type 2 diabetic rats,up-regulated the expression of GluT4,promotes the oxidation of glucose in the liver,but also increases the uptake of glucose in the cell,reduces blood sugar and improves the sensitivity of insulin.
