Objectiv: To investigate the effect of RvD1 on toll-like receptor 4(TLR4)and glucose transporter 4(GluT-4)in skeletal muscle of type 2 diabetic mice.Methods: Thirty five male C57 BL / 6 mice were randomly divided into control group,type 2 diabetes mellitus group and prolactin group.The mice in the control group were given normal diet diet,the mice in diabetes mellitus group and prolactin group were given high glucose and high fat diet,free drinking water and eating.After 6 weeks,streptozotocin(STZ,100 mg / kg)was injected intraperitoneally with type 2 diabetes mellitus and RvD1 group.After the success of the model,the control group of mice free feeding and drinking water;diabetic mice with intraperitoneal injection of PBS solution 0.2ml every day;RvD1 mice with intraperitoneal injection of RvD1 0.2ml every day.Continuous feeding 21 days to detect blood lipids,serum insulin levels and inflammatory factors,take muscle tissue to detect related indicators.Results:1 Body weight change in mice:After 6 weeks of high-sugar and high-fat diet,the body weight of the mice was higher than that of the common diet group.After the model was successful,the body weight of the mice in T2 DM group increased.Elimination of RvD1 for 2 weeks,compared with T2 DM group,T2DM+RvD1 group mice weight loss.2 Changes of blood lipid levels in miceThe levels of cholesterol and triglyceride in T2 DM group were significantly higher than those in NC group(P <0.05).The level of cholesterol and triglyceride in T2 DM + RvD1 group was significantly lower than that of T2 DM group(P <0.05).3 Changes of fasti ng blood gl ucose and i nsuli n resist ance i ndex i n mi ceThe level of fasting blood glucose in the high-sugar and high-fat diet group was higher than that in the normal diet group(P <0.05).After 3 weeks of RvD1 intervention,the blood glucose and insulin resistance index of T2DM+RvD1 group were lower than those of T2 DM group(P <0.05).4 Inflammatory factor levels in miceCompared with the NC group,the levels of IL-6 and TNF-? in T2 DM group increased,the difference was statistically significant(P <0.05),and the IL-6 and TNF-? in T2 DM +RvD1 group were decreased(P <0.05).5 TLR4 changes in miceThe level of TLR4 in T2 DM group mice was higher than that in NC group(P <0.05),while that of T2 DM +RvD1 group was lower than that in T2 DM group(P <0.05).6 GluT4 mRNA expression level of miceThe mRNA expression level of GluT4 in mice was detected by PCR,and the mRNA expression of GluT4 in T2 DM group was lower than that in NC group(P <0.05);the expression of GluT4 mRNA in T2DM+RvD1 group was significantly higher than that in T2 DM group(P <0.05).Conclusions: Appropriate amount of RvD1 can decrease the body weight,blood glucose,insulin resistance index,blood lipid and inflammatory factor level in T2 DM group mice,and reduce the level of TLR4 and GluT-4 mRNA expression in skeletal muscle in T2 DM group mice,thereby improving insulin resistance.
