| Purpose:The aim of this study was to investigate whether plasma miR-21 and let-7i levels in patients with advanced gastric cancer were different from those in healthy controls,to explore the correlation between plasma miR-21,let-7i expression and chemotherapy efficacy and disease progression in XELOX regimen。 Methods: Sixty 60 cases of early diagnosis and treatment of advanced gastric cancer patients were treated with XELOX regimen,and the curative effect was evaluated every 2 cycles after chemotherapy.No progress of the disease was continued.Before chemotherapy,2 cycles after chemotherapy,When the disease progresses,plasma samples are prepared.Plasma samples of 60 healthy controls were prepared in the same period.The expression of miR-21 and let-7i was detected by PCR in the plasma samples.The expression of miR-21,let-7i and its correlation with chemotherapy efficacy and disease progression were analyzed by statistical analysis.Result:1.The expression of miR-21 in the patients with advanced gastric cancer was significantly higher than that in the control group,the difference was statistically significant(P<0.01).The expression of let-7i in patients with advanced gastric cancer and healthy control group was not significantly changed,the difference was not statistically significant(P= 0.782).2.The expression of miR-21 was significantly down-regulated in patients with advanced gastric cancer before and after chemotherapy,the difference was statistically significant(P<0.01);let-7i had no significant change before and after chemotherapy,the difference was not statistically significant(P= 0.568).3.The expression of miR-21 was significantly down-regulated in patients with advanced gastric cancer,and the difference was statistically significant(P <0.01).The expression of miR-21 was not significantbefore and after chemotherapy in patients with advanced gastric cancer,the difference was not statistically significant(P =0.54).The relative expression of miR-21 in patients with chemotherapy was significantly higher than that in the control group(P<0.01).4.The relative expression level of miR-21 in effective chemotherapy group was higher than that before chemotherapy(P<0.01),and higher than that after chemotherapy(P<0.01).The relative expression of plasma in invalid chemotherapy group miR-21 was higher than that before chemotherapy(P<0.01),and higher than that after chemotherapy(P<0.01).Conclusions: Plasma mi R-21 levels may be useful in predicting the efficacy and progression of XELOX chemotherapy in patients with advanced gastric cancer. |
