Sjogren's syndrome(SS)is a common autoimmune disease,which m ainly involves exocrine glands,especially in lacrimal glands and salivar y glands.Dry mouth,dry eye are the main clinical manifestations.How ever,the disease often extend beyond the exocrine glands to seriously a ffect other organs systems,inducing multiple system damage.But its pa thogenesis is not clear.It is believed that abnormal activation of T and B lymphocytes plays an important role.It has been shown that the expr ession of p-STAT3 and SOCS1 in peripheral blood lymphocytes of pati ents with systemic lupus erythematosus(SLE)has been changed.,if per ipheral blood lymphocytes stimulatied by the cytokine IL-21,which det ermines the function of Tfh cells.Other studies have shown that the ex pression level of p-STAT3 in the synovium of rheumatoid arthritis rats is increased.It is suggested that p-STAT3 plays an important role in so me autoimmune diseases,and the expression of p-STAT3 in cytokine-in ducible inhibitors of signaling is very important for the expression of S OCS1.And there are few studies about the role of p-STAT3 and SOCS1in the activation of T lymphocytes and the apoptosis of labial gland e pithelial cells.Objective: In this study we collected the patients' salivary glands with clinically diagnosed primary Sjogren's syndrome.Then we measured the expression levels of p-STAT3 and SOCS1 in different pathological grades of labial glands by immunofluorescence and Western Blot method.Compared the expression differences between the grade?,? group and the grade III,IV group,further observed the differences between these two kinds of protein expression between class III and IV pathological group of patients,and to explore its relationship with CD4~+ T cells.The aim was to confirm whether thep-STAT3/SOCS1 signaling pathway through activation of T cells inducing takeplacement and development of Sjogren's syndrome,to provide new targets for the future treatment of Sjogren's syndrome.Methods:1 By single standard immunofluorescence and Western blot,we detecte the expression rate of p-STAT3 and SOCS1 positive cells and the relative protein expression in the labial glands of the grade?,? group and the grade III,IV group.Double labeled immunofluorescence assay was used to detect the expression of p-STAT3 and SOCS1 in CD4~+T cells in the labial glands of the grade?,? group and the grade III,IV group.2 Statistical analysis: SPSS21.0 statistical software was used for statistical analysis,the positive number of patients in the grade?,? group and the grade III,IV group were compared by Fisher exact probability test,P<0.05 was statistically significant.The comparison of the positive cell rates between the grade III and IV groups was compared by rank sum test,and P<0.05 was statistically significant.Results :1 The expression of p-STAT3 and SOCS1 in labial gland tissue detected by immunofluorescence single labelingThe results of immunofluorescence single labeling showed that more p-STAT3 green fluorescent particles were found in the lymphocytes of labial glands of the grade III,IV group with Sjogren's syndrome,while only a small amount of green fluorescent protein expressed in the grade?,? group.At the same time,more SOCS1 green fluorescent particles were also found in the lymphocytes of labial glands of the grade III,IV group,while only a small amount of green fluorescent protein expressed in the grade?,? group.In addition,the expression of p-STAT3 and SOCS1 was also found in the epithelial cells of labial gland duct.2 The expression of p-STAT3 and SOCS1 in labial gland tissue detected by Western blotWestern blot analysis showed that,compared with the grade?,?group(0.083±0.063),the expression of p-STAT3 protein in the labial glands of the grade III,IV group was increased,and the expression level of p-STAT3 in the ? grade(2.777±0.117)labial glands was higher than that of the grade ?lesions(1.089±0.001).At the same time,the expression level of SOCS1 protein was higher than that of the grade?,? group(0.033±0.002),and the expression level of grade ? lesions(1.233±0.051)was higher than that of grade ?(0.506±0.011).3 Detection of expression of CD4~+T cells in labial gland by immunofluorescence single labelingThe results of immunofluorescence single labeling showed that a large number of CD4~+ red fluorescent particles were found in the lymphocytes of labial glands of the grade III,IV group with Sjogren's syndrome.4 The expression of p-STAT3 and SOCS1 in CD4~+T cells by immunofluorescence double labeling detectionCD4/p-STAT3 double immunofluorescence showed that,in CD4~+T lymphocytes which is in focal lymphocytic of the labial gland tissue of grade III and IV of Sjogren's syndrome,can we see the expression of p-STAT3,and the positive rate of ? patients with lesions in CD4~+p-STAT3+T lymphocytes is higher than the ? level group,no significant difference of the expression between the two groups by statistical analysis.(Z=-0.645,P=0.519,P>0.05).CD4/SOCS1 double immunofluorescence showed that,in CD4~+T lymphocytes which is in focal lymphocytic of the labial gland tissue of grade III and IV of Sjogren's syndrome,can we see the expression of SOCS1,and the positive rate of ? patients with lesions in CD4~+SOCS1+T lymphocytes is higher than the ? level group,no significant difference of the expression between the two groups by statistical analysis.(Z=-0.645,P=0.519,P>0.05).Conclusions:p-STAT3/SOCS1 protein may induce injury of labial gland tissue in Sjogren's syndrome by activating CD4~+T cells,and whether it accelerates the progression of the disease and the severity of the lesions of the labial gland remains to be further confirmed.
