Objective:To study the CD127's expression in labial gland tissue of patients with p SS and analysis the correlation between the expression and other disease activity indicators of the patients with p SS,to explore the relationship of CD127 and the pathogenesis of p SS,in order to find a certain regularity,further guide clinical work.Methods:In the experimental group,there were 30 patients with primary sjogren's syndrome in the international classification(diagnosis)which was revised in 2002.In the control group,there were 10 cases of normal labial gland tissue removed from the surgical incision of the lip of patients with mucous cyst or lip trauma..Two groups above were collected,and HE staining was used to confirm the pathological diagnosis.Immunohistochemical experiments were conducted to locate the expression sites of CD127,and the protein expression level of CD127 was partially quantitatively analyzed by positive cell count method.Extract m RNA and reverse transcription-polymerase chain reaction(rt-pcr)was used to detect the m RNA expression level of CD127.The expression levels of CD127 in the experimental group and the control group were compared,and the relationship between CD127 expression level and Chisholm grading and other disease activity indicators of p SS patients was statistically analyzed,and the results were summarized.Results:1.Immunohistochemical staining results showed that the experimental group CD127 was uniformly expressed in infiltrating lymphocytes around the ducts,targeting cytoplasm or cell membrane,and the control group had little or no expression of CD127.The expression level of CD127 protein in the experimental group(3.03±0.96)was significantly higher than that in the control group(0.30±0.67),and the difference was statistically significant(P<0.01).2.Rt-pcr results showed that the expression level of CD127 m RNA in the experimental group(5.32±4.50)was significantly higher than that in the control group(0.40±0.53),and the difference was statistically significant(P<0.01).3.CD127 expression level was significantly positively correlated with the Chisholm grading results(P<0.01).4.The expression level of CD127 was not significantly correlated with the patients' subjective mouth dry months,Schirmer, antibodies and ECT results(P>0.05).Conclusion:In labial gland tissues of patients with primary sjogren's syndrome(pSS),CD127 m RNA and protein expression levels are higher than normal control group,and is significantly positively related to the severity of lymphocyte infiltration of gland.It is speculated that CD127 is involved in the primary sjogren's syndrome.
