Objectives By observing the effect of acupuncture on the expression of Col I,FN and VEGF in lung tissue of bronchial asthma rats,to clarify the relationship between Col I,FN,VEGF and asthma and the regulation of acupuncture.Methods 90 SPF male SD rats were randomly divided into normal control group,asthma model group,and acupuncture treatment group,30 rats in each group.The asthmatic models were induced by intraperitoneal injection of OVA and Al(OH)3 mixture sensitized and aerosolized inhaled OVA solution.The normal control group was inhaled with saline,and the acupuncture treatment group was inhaled OVA solution after acupuncture.According to the difference of total intervention time,10 rats in each group were sacrificed by random number table after 2,4 weeks,and the remaining rats were sacrificed 6 weeks after atomization.We marked the rats of aerosolized 2,4,6 weeks of normal control group as A,B,C,the rats of aerosolized 2,4,6 weeks of asthma model group as D,E,F,the rats of aerosolized 2,4,6 weeks of acupuncture treatment group as G,H,I.The lung tissue morphology and airway remodeling situation of each group rats observed by HE staining and Masson staining,the expression of Col I,FN and VEGF in lung tissue of each group rats were detected by immunohistochemistry method and ELISA.Results 1 The rats of asthma group appeared nervous,quick and deep breath,cyanotic mouth and incontinence after accepting the agonist atomization,the symptom of the rats in acupuncture group and control group during atomization is lighter.2 HE staining showed that the lung tissue of the rats in asthma group appeared serious airway remodeling and inflammation of the airway pathologic changes,and with the prolongation of atomization cycle,the pathological changes of lung tissue in D,E,F group increased.Acupuncture group and the control group had no obvious pathological changes in lung tissue of rats,and the lung tissue structure had no obvious change with the prolongation of atomization cycle.3 Masson staining showed that the lung tissue of rats in asthma group increased in values of WA/Pi,WAm/Pi,WAc/Pi,WAi/Pi compared with the blank group(P<0.05),as the growth of the atomization cycle,there was a growing trend in the average value of WA/Pi,WAm/Pi in D,E,F group,WAi/Pi appeared no thickening phenomenon with the prolongation of atomization cycle(P>0.05).The lung tissue of rats in acupuncture group and the blank group,WA/Pi,WAm/Pi,WAc/Pi,WAi/Pi appeared no significant difference(P>0.05),4 The Col I,FN,VEGF expression of lung tissue of rats in asthma group was obviously higher than that of control group according to the detection of immunohistochemical method and ELISA method(P<0.05),and with the extension of atomization cycle,the Col I,FN,VEGF expression of D,E,F group appeared downward trend,but still higher than the control group,the difference has statistically significance.The expression of Col I,FN,VEGF in the lung tissue of rats in acupuncture group was lower than that in the asthma group(P<0.05),the difference has statistically significance.Conclusions 1 Acupuncture can significantly improve breathing difficulties,cyanotic mouth,nervous and other symptoms of asthma rats by the atomization stimulation.2 As the times of atomization increased,the degree of airway remodeling was increased in model group D,E and F,will acupuncture can obviously inhibit the occurrence of airway remodeling development of asthmatic rat.3 The expression of Col I,FN,VEGF in lung tissue of asthma model group rats increased,and as the times of atomization increased,the expression of Col I,FN,VEGF in lung tissue of asthma model group rats appeared downward trend.4 Acupuncture can inhibit Col I,FN,and VEGF expression of lung tissue of asthma rat.
