Objective:In this study,we detected the expression levels of HIF-1α,VEGF,TGF-β1 in serum of patients with asthma attack and remission and healthy groups,and observed the correlation between the levels of HIF-1α,VEGF,TGF-β1 and the pathogenesis of bronchial asthma to discuss the role and mechanism of vascular factors in the pathogenesis of bronchial asthma.Methods:From August 2020 to February 2021,30 patients in the respiratory and critical care department of our hospital who is hospitalized or outpatient with acute asthma who met the inclusion criteria were collected as experimental group,and 15 patients with asthma in remission meeting the inclusion criteria as the remission group,15 healthy subjects in our hospital were collected as the control group during the same period.There was no statistical difference in age and gender composition among the three groups(P>0.05).Fasting blood samples were collected from all subjects,including inpatients on the morning after admission,outpatients on the day of admission,and healthy subjects on the day of physical examination.Serum samples were centrifuged and collected,and stored in a refrigerator at-70℃.Then detect the serum levels of HIF-1α,VEGF and TGF-β1 by enzyme-linked immunosorbent assay(ELISA).At the same time,pulmonary function,blood gas analysis,blood routine and allergen measurement data of patients in asthma attack and remission stage were collected.Blood routine data of healthy group were collected.Then record the FEV1%pred,FEV1/FVC value,oxygen partial pressure,eosinophil count and IgE value.Related data were statistically analyzed.Then analyze the data.Results:1.The levels of serum HIF-1α,VEGF and TGF-β1 in asthma attack group were significantly higher than those in remission group and healthy group,with statistical significance(P < 0.05).The levels of serum HIF-1α,VEGF and TGF-β1 in remission group were significantly higher than those in healthy group,with statistical significance(P < 0.05).2.Serum HIF-1α was positively correlated with VEGF and TGF-β1(P < 0.05),and serum VEGF was positively correlated with TGF-β1(P < 0.05).3.The partial pressure of oxygen,FEV1%pred and FEV1/FVC in the attack group were significantly lower than those in the remission group,the difference being statistically significant(P < 0.05);the peripheral blood eosinophil count and IgE value in the attack group were significantly higher than those in the remission group,the difference being statistically significant(P < 0.05).4.The levels of HIF-1α,VEGF and TGF-β1 in asthmatic patients decreased with the increase of partial oxygen pressure,and the partial oxygen pressure was negatively correlated with the levels of HIF-1α,VEGF and TGF-β1(P < 0.05).5.Eosinophil count was negatively correlated with oxygen partial pressure,FEV1%pred,FEV1/FVC(P < 0.05),and positively correlated with IgE,HIF-1α,VEGF,TGF-β1 levels(P < 0.05).6.FEV1%pred was significantly correlated with the expression levels of serum HIF-1α,VEGF and TGF-β1 in patients with asthma(P < 0.05).Conclusion:1、The levels of HIF-1α,VEGF,and TGF-β1 in patients with asthma at attack stage were significantly higher than those in remission stage and healthy group,and there was a positive correlation among the indicators.Asthma attack was correlated with the increase of HIF-1α,VEGF,and TGF-β1,and blood vessels were involved in the process of asthma inflammation and airway remodeling.2、The peripheral blood eosinophil count at the attack stage was higher than that at the relief stage,and was significantly correlated with the levels of FEV1%pred,FEV1/FVC,IgE,HIF-1α,VEGF,TGF-β1.The eosinophil count,together with HIF-1α,VEGF,and TGF-β1,increased the airway inflammation of asthma.3、FEV1%Pred was negatively correlated with HIF-1α,VEGF and TGF-β1.The higher the expression level of HIF-1α,VEGF and TGF-β1 in serum,the lower FEV1%Pred was,the worse the lung function was,and the more serious the disease was.
