Objective:To explore the characteristic of women's urinay tract bcterial communities,we use the un-cultivated method of 16 S r RNA gene high-throughput sequencing to contrast two groups of women,the members of experimental group are menopausal women who have the symptoms of frequent urination?urgent urination and dysuria,the members of control group are menopausal women who are healthy.Methods:Collecting the urine specimen of 14 menopausal women from Affiliated Hospital of Qingdao University Nephrology outpatient who had the symptoms of frequent urination?urgent urination and dysuria,each urine sample was divided into two,one was taken to extract the bacterial genomic DNA,then we detect the diversity of the urine samples by using the 16 S r RNA gene high-throughput sequencing technology,the other was taken to the clinical bacteriology laboratories.Collecting the urine specimen of 14 menopausal women from Affiliated Hospital of Qingdao University Physical examination center who are healthy,in the same way,we also detect the diversity of these urine samples by using the 16 S r RNA gene high-throughput sequencing technology.We contrast the characteristic of the urinary tract bcterial communities between the two groups.Results:(1)The sequencing results showed that diversity and kinds of orgnisms of urinry tract varied among women.The sequences from all the two groups urine samples were classified into 385 genera,each sample has at least 25 genera on average.The most abundant phyla detected were firmicutes?Actinobacteria,Proteobacteria and Bacteroidetes.At the genus level,the most abundant members were Lactobacillus,Gardnerella,Corynebacterium,Enterobacteriaceae,Staphylococcus,Streptococcus,Anaeroc occus,Peptoniphilus,Prevotella and Actinomyces.(2)Compaired to the control group,the experimental group has a higher positive rates of Gardnerella?Aerococcus,and a lower positove rate of Lactobacillus.The difference is statistically significant.(P?0.05).Bacteria such as Enterobacteriaceae,Anaerococcus,Actinobaculum,Actinomyces,Sneathia,Prevotella and Oligella were more in the experimental group than in the control group.Other bacteria such as Streptococcus,Rhodanobacter,Peptoniphilus and Bifidobacterium were more in the control group than in the experimental group,for the reason that the number of our specimen was too smll,the above two differences had no statistically significant(P?0.05).Conclusions:The urine microbiome of menopausal women presented diverse.The most popular bacteria are Firmicutes,Actinobacteria,Proteobactria,and Bacteriodet.Of all the two group specimen,the most popular dominant bacteria is Lactobacillus,the second most popular dominant bacteria is Gardnerella.The urinary tract bcterial communities of menopausal women who have the symptoms of frequent urination,urgent urination and dysuria has a higher positive rates of Gardnerella,Aerococcus Actinobaculum and Sneathia,and a lower positove rate of Lactobacillus.It reveals that menopausal women with the urinary tract infection symptoms has a different urinary tract bcterial communities structure,and it is likely that the difference is an important reason for why the incident rate of menopausal women urinary tract infection is higher.
