The Effects Of Hydroxy Fasudil (HF) On Aortal Endothelial And The Expression Of IL-8?TGF-?1 After Balloon Endothelial Denudation Of Aorta In Rats

Objective:To investigate the effects of HF on aorta endothelial after balloon injury and the concentration of IL-8?TGF-?1 in plasma and in neointima and artery-medial in rats.Methods:Forty sale SD rats were divided into three groups,the control group(n=16),the operation group(n=16,balloon endothelial denudation with NS intrapreitoneal injection0.5ml/100g/bid for 28d),the HF group(n=16,balloon endothelial denudation supplemented with fasudil intrapreitoneal injection 0.5mg/100g/bid for 28d).Then the rats aorta tissue were taken after 14d?28d from the three groups,and the morphological characteristics were observed by light microscope.The tissue sections from the three groups,the aortic intimal area(IA)?medial area(MA)?medial thickness(MT)?intimal thickness(IT)?(IA/MA)were measured after 14 d and 28 d.And compared with the serum level of IL-8?TGF-?1 taken from renal venous from the rats.The expression of TGF-?1 in neointima and artery-medial were measured in operation group and HF group.Results : only a flat endothelial cells arranged in normal intimal,the middle laye was spindle-shaped smooth muscle cells.In the operation group after 14 ds,the intimal hyperplasia,the increased values of the aortic intimal area(IA)?medial area(MA)?medial thickness(MT)?intimal thickness(IT)?(IA/MA)were significant compared with control group(p<0.05).The difference of the above measured values was continuing to 28d(p<0.01).The measure values of IA?MA?IT?MT?IA/MA between the HF group and operation group were no difference(p>0.05),but 28 ds later the above measured values in HF group were improved compared with the operation group(p<0.05).The concentration of IL-8?TGF-?1 in plasma and the expression of TGF-?1 in neointima and artery-medial was increased in the two balloon injury groups than the control group after 14 d later(P<0.01),but the measured values in HF groups were less than in operation groups in plasma(p<0.05).The concentration of IL-8?TGF-?1 in plasma and TGF-?1 in neointima and artery-medial in HF group were decreased significant than those in the operation group after 28d(p<0.05).Conclusion;The results indicated that hydroxy fasudil can efficiently inhibits intimal proliferation during restenosis after balloon injury of aorta in rats.It may be related to the early inhibition of IL-8?TGF-?1.
