[Objective] To study the effects of fasudil combining with Rosiglitazone on the expression of VCAM-1 (vascular cell adhesion molecule-1)and MCP-1 (monocyte chemotactic protein)in HCAEC(human coronary arterial endothelial cells) induced by lipopolysaccharide (LPS).[Methods] we conducted an experiment using the 3rd to the 5th generation of HCAEC lines in vitro.group of experiments:(1)Normal group (non-intervention); (2)Lps group (100ng/ml); (3)Fasudil group (10nM); (4)Rosiglitazone group (10ug/ml);(5) LPS+ Fasudil group(10nM Fasudil+ 100ng/ml LPS); (6) LPS+Fasudil+ Rosiglitazone group (lOnM Fasudil+10ug /ml rosiglitazone+ 100ng/ml LPS); The expression of the sVCAM-1 and MCP-1 protein in supernatant were detected by ELISA and their mRNA were detected by RT-PCR.[Results] Compared with the normal group, the expression of VCAM-1,MCP-1mRNA and sVCAM-1,MCP-lprotein were enhanced by LPS (P<0.05). In basal conditions, Fasudil and Rosiglitazone had no significant effect on VCAM-1, MCP-1 mRNA and sVCAM-1, MCP-1 protein expression (P>0.05),but Fasudil inhibits the expression of MCP-1, sVCAM-1 protein and VCAM-1、MCP-1 mRNA as well which induced by LPS (P<0.05), compared with Group(5), the Group(6) significantly inhibited LPS-induced VCAM-1, MCP-1 mRNA expression (P<0.05), as well as the MCP-1, sVCAM-1 protein expression (P<0.05).[Conclusion] Fasudil inhibits the expression of MCP-1, VCAM-1 in the coronary artery endothelial cells especially combined with Rosiglitazone.
