In this experiment,peanut meal was used as the main raw material and the Bacillus natto was used as the fermentative strain.The extract of the peanut meal fermentation,were used to study the antioxidant activity,the anti-aging effect on the female reproductive system,and the regulation role of intestinal flora in mice.Experiment 1: anti-aging effect of peanut meal fermentation on reproductive function of female ratsA female aging rats model was established.72 female SD rats were weighted and randomly divided into groups according to the area of body weight: the control group N?normal saline?,the model group M?normal saline?,the low dose group?FL,25 times dilution of fermentation?,the middle dose group?FM,5 times dilution of fermentation?,the high dose group?FH,fermentation substance stock solution?,and the positive control group Y?VE100mg/kg?.There were twelve rats in each group.Intraperitoneal injection of D-galactose?400 mg / kg.d?were performed in each group except the intraperitoneal injection of saline was performed in the blank group.The intragastric dose of each group was 0.2 ml/10 g.d.After 8 weeks of experiment,animals were sacrificed,blood and organs were collected,the organ index was calculated,the histological observation was performed,and the indexes of antioxidation and sex hormone were determined.The uterine index and ovarian index in the FM group and the Y group were significantly higher than those in the M group?P<0.05?.The results of the determination of the serum E2 showed that the expression level of E2 in the FM group and the FH group were higher than the M group?P<0.05?.The expression levels of serum FSH in the FM group,the FH group and the Y group were significantly lower than that in the M group?P<0.05?.The expression levels of serum LH in each treatment group were significantly lower than those in the M group,and there was significant difference between the two groups?P<0.05?.The results showed that peanut meal fermentation had antioxidant activity in vivo.The medium and high dose of HFY can delay the aging of the female reproductive system.Experiment 2: the effect of peanut meal fermentation on intestinal function of antibiotic-induced diarrhea in miceThe mouse diarrhea model was established.40 Kunming mice were weighted and randomly divided into the blank group A?saline?,the model group B?ampicillin?,the natural recovery group C?ampicillin + saline?and the HFY group D?ampicillin +HFY?.In addition to the blank group?A?,the treatment group were given to diarrhea with the ampicillin dose was 2 mg / g bw.d,which is equivalent to 5 times the adult dose,and the model was established.The mice of model group?B?were sacrificed and the intestinal contents were collected.In addition to the control group?A?and the natural recovery group?C?which were given to normal saline,the HFY group?D?was given to the original fermentation broth with the dose of 0.2 ml/10 g.d,the mice were sacrificed 5 days after intragastric administration,and the intestinal contents were collected.Based on the Illumina Hi Seq2500 sequencing platform,high throughput sequencing of intestinal contents was performed to explore the classification and abundance analysis of intestinal flora.The results showed that at the division level,the top three species of the group were Bacteroidetes,Firmicutes,and Proteobacteria,The relative content of Bacteroidetes were 79.87% in the group B 65.90% in the group C,58.22% in the group A and 45.10% in the group.The order of each group on the level of Firmicutes was D,A,C,B from high to low.The relative content of Proteobacteria in each group was higher in the group B?17.73%?and the group C?17.60%?than the group A?11.63%?,and the group D?6.92%?.At the genus level,the group B showed a low level except for escherichia-shigella.LachnospiraceaeNK4A136group of group D was higher than the other groups;Escherichia-Shigella of group A and group D were lower than those in group B;Roseburia of group D was higher than the other groups;Clostridiumsensustricto1 of group D higher than in B group;Coprococcus1 of A and D were higher than group B;group D was higher than that of Lactobacillus of A,B two groups;group D was higher than that of Enterococcus of A.Tyzzerella,Lachnoclostridium and LachnospiraceaeUCG-010 of D group were higher than that of group C.Species observed,Shannon,Chao1,ACE,PDwholetree index of B group were lower.The diversity of the diversity index showed that there was a significant difference between the group B and other groups?P<0.05?.Based on the Shannon index analysis,the species richness of the group D was higher than that of the other three groups?P<0.05?.The results showed that peanut meal fermentation on diarrhea mice intestinal bacteria can play a regulatory role,increase the number of beneficial bacteria,inhibit the growth of harmful bacteria,increase the intestinal flora richness and species evenness.The composition of the intestinal flora and the flora of the diarrhea mice were better than that of the natural recovery group and the blank group.
