Based On High-throughput Sequencing To Explore The Effect Of Shenlingbaizhusan On The Intestinal Flora Of Diarrhea Due To Deficiency Of Spleen-qi

Objection: A stable intestinal micro-ecological environment plays an important role in human health.Many diseases can be traced back to the imbalance of intestinal flora,and the occurrence of functional diarrhea is also closely related.However,previous studies on the dysbacteriosis of intestinal tract in functional diarrhea have been limited to the means of bacterial culture,and it is difficult to obtain more specific changes in flora.The emergence of high-throughput sequencing methods fills this gap.This study hopes to explore the effects of functional diarrhea on intestinal microflora at different biological levels through high-throughput sequencing,and to clarify the effects of specific microflora on functional diarrhea,and to explore the function of Shenling Baizhu Powder.Regulation of the specific flora of the intestinal tract of sexual diarrhea.This paper mainly discusses the understanding and research of functional diarrhea from two parts.The first part is the literature review of TCM literature,the meta-analysis of the effect of Shenling Baizhu Powder on functional diarrhea and the review of Western medical literature.The review of Chinese and Western medicine literature mainly involves the etiology,pathogenesis,TCM treatment and Western medicine of Chinese medicine.Advances in modern clinical research on diarrhea,including etiology,pathogenesis,and treatment.The metaanalysis of Shenling Baizhu Powder in the treatment of functional diarrhea explores the therapeutic effect of Shenling Baizhu Powder on functional diarrhea through literature search,analysis and data integration.The second part is to explore the changes of flora from clinical experiments.The effects of sexual diarrhea and the regulation of intestinal flora by Shenling Baizhu Powder.Method: This study collected 20 patients from the spleen and stomach clinic of Dongzhimen Hospital of Beijing University of Chinese Medicine from January to December in 2018.The patients diagnosed as functional diarrhea and TCM syndrome was a spleen deficiency.The randomized grouping method was used,and the experimental components were the traditional Chinese medicine group(n=10)and the blank control group(n=10).The healthy control group selected 20 volunteers who were excluded from other medical history by physical examination and laboratory examination in the physical examination center of the external hospital during the time period.The changes of clinical symptom scores before and after treatment in the Chinese medicine group were observed.At the same time,the fecal samples from the experimental group and the fecal samples from the healthy control group were collected for high-throughput sequencing to explore the effect of specific flora on functional diarrhea.The fecal samples from the traditional Chinese medicine group and the blank control group were collected before and after treatment to explore Shenling Baizhu Powder.For the influence of intestinal flora,the effect of self-regulation on intestinal flora is also excluded.Results: 1.Among the experimental group,there were 11 people without anxiety or depression,3 people with anxiety or depression,and 6 people with obvious anxiety or depression.In the healthy control group,there were 18 people without anxiety or depression,which may be anxiety or depression.2 people,0 people with obvious anxiety or depression.The data of the two groups were accurately tested by Fisher,?2=7.992,P=0.014(27)0.05..2.After treatment with traditional Chinese medicine,0 cases were Coprococcus and Megasphaera in the experimental group was higher than that in the healthy control group.4.For the Chinese medicine group and the blank control group before and after treatment,both groups had changes in Lactobacillaceae,but the changes in the Chinese medicine group were more obvious.Based on the clinical symptoms and intestinal flora before and after treatment in the Chinese medicine group,Shenling Baizhu Powder may improve the clinical symptoms of patients with spleen-deficiency type diarrhea by adjusting the number of Lactobacillaceae(Lactobacillaceae).Conclusion: 1.There was a significant difference in mood between patients with functional diarrhea and healthy controls,indicating that anxiety and depression were closely related to the occurrence of functional diarrhea;2.Comparing the changes of clinical symptom scores before and after treatment,it indicated that Shenling Baizhu Powder has a good therapeutic effect on spleen deficiency and diarrhea.3.Based on high-throughput sequencing,it can be found that there is no significant difference in the abundance and quantity of intestinal flora between patients with functional diarrhea and healthy people,but there are still differences in the composition of the flora.The increase of different species of different species Veillonellaceae,Erysipelotrichaceae,Coprococcus,Megasphaera,Erysipelotrichi,Coriobacteriia,Mollicutes,the reduction of Synergistetes,Synergistaceae,Dethiosulfovibrionaceae may be related to the occurrence of functional diarrhea.4.Comparing the changes of the flora before and after the Chinese medicine group and the blank control group,it indicates that the human body has certain self-regulation ability in the intestinal flora,and Shenling Baizhu Powder has a regulating effect on the intestinal flora.Shenling Baizhu Powder may improve the clinical symptoms of patients with spleen-deficiency type diarrhea by adjusting the amount of Lactobacillaceae.
