Objective: To observe the expression change of c-IAP-1 and Caspas-3,-7 in brain tissue around the experimental hematoma and the effect of statin intervention,to explore the possible neuroprotective effect of statins on cerebral hemorrhage and its mechanism in order to play a guiding role in clinical medication.Methods:(1)60 SD rats were randomly divided into sham operation group(20 rats),ICH group(20 rats)and rosuvastatin group(20 rats).Each group was divided into four time points,6h,24 h,72h and 5d,each time point for 5 rats.ICH Model was established by injecting autologous arterial blood into brain tissue.(2)All the rats were scored on the Neurological deficit by Garcia.(3)We observe the sectioned and stained brain tissue by microscope.(4)Finally we tested the expression of c-IAP-1 and Caspase-3,-7 by RT-qPCR four time point after the operation.Results:(1)The Garcia score of the cerebral hemorrhage group and the rosuvastatin intervention group at the same time point showed that the neurological deficit in the rosuvastatin intervention group was lower than that in the cerebral hemorrhage group(P <0.05),the neurological function of the cerebral hemorrhage model group was significantly higher than that of rosuvastatin group,the statistical significance exists.(P <0.05).(2)c-IAP-1 and Caspase-3,-7 were significantly increased by immunohistochemistry after intracerebral hemorrhage.After 6 hour of cerebral hemorrhage,the positive expression of c-IAP-1 and Caspase-3,-7 was observed in the brain tissue around the hematoma.The expression of Caspase-3,-7 in the rosuvastatin group(group C)was significantly lower than that in the cerebral hemorrhage model group(group B)at the other time points except for 6 hours of cerebral hemorrhage The Rosuvastatin significantly reduced Caspase-3,-7 and increased c-IAP-1 levels.(3)Analysis by RT-qPCR,compared with cerebral hemorrhage model group and rosuvastatin group,the expression of c-IAP-1 and Caspase-3,-7 in false surgical group was lower.Expect the 6h,the expression of c-IAP-1 was higher and the expression of Caspase-3,-7 was lower in cerebral hemorrhage model group,and the differences was significant(P < 0.05).Conclusion:(1)Rosuvastatin can reduce the neurological deficit after cerebral hemorrhage,and can product the nerve function.(2)Rosuvastatin can inhibit the activation of Caspase-3 and Caspase-7 to impove the level of c-IAP-1,c-IAP-1 inhibit the expression of Caspase-3 and Caspase-7 reduce the cell stress reaction of cerebral hemorrhage,and reduce the secondary brain damage affected by induced apoptotic response,play the role of nerve protection function.
