Objective1.To explore an efficient method to develop oral-breathing model in mice.2.To establish oral-breathing model based on Senescence-accelerated mouse prone(SAMP)and detect the mitochondrial dysfunction.Methods1.A total of twenty C57BL/6 mouse were divided into suture group(n=10)and packing group(n=10).Mouse in suture group would accept nostril suture while mouse in packing group would accept nasal packing.The motality and ratio of nasal occlusion lost in two groups would collected for a further analysis.2.A total of 12 SAMP6 mice and 12 SAMR1 mice with nasal packing were made out to oral-breathing model(SAMP6_OR/SAMR1_OR).Then we raised them to age of 4month(n=4),6 month(n=4),8 month(n=4).The same number of coetaneous SAMP6 and SAMR1 mice were matched for study.All mice' tissues such as skin,upper airway muscle,musculus rectus abdominis,genioglossus,buttock muscle and leukocytes were collected for further samples making.Mitochondrial membrane potentials(MMP)were detected by flow cytometry(FCM)after JC-1 loading.Further data analysis was performed.Results1.Comprasion of motarlity and the ratio of unstability:1)Motarlity of nostril suture group(70%)was significantly greater than nasal packing group(0%)(P<0.05).2)There was no significance in the falling off ratio betweent suture group(50%)and nasal packing group(20%)(P>0.05).2.Study of mitochondrial dysfunction in oral-breathing model based on SAMP mouse:1)MMPs(Mitochondrial Membrane Potentials)in majority parts of tissues in SAMP6 were less than in SAMR1(P<0.05).2)MMPs of skin(P<0.05),upper airway muscle(P<0.05),musculus rectus abdominis(P<0.05),upper airway mucosa(P<0.05),genioglossus(P<0.05),buttock muscle(P<0.05)and leukocytes(P<0.05)in SAMP6 had statistical significanse among 4-month-old,6-month-old and 8-month-old group.3)MMPs of upper airway mucosa(P<0.05),genioglossus(P<0.05)and leukocytes(P<0.05)in SAMR1 had statistical significanse among 4-month-old,6-month-old and 8-month-old group.There were no significance among 4month-old,6 month-old and 8 month old group in MMPs of skin(P>0.05),upper airway muscle(P>0.05),musculus rectus abdominis(P>0.05)and buttock muscle(P>0.05).Conclusions1.The nasal packing method to establish oral-breathing model in mice is better than the method of nostril suture in asecpts of less truma,low motarlity and stability.2.Mitochondria function is negatively related with age both in senescence-accelerated mouse and control.The aging processing in different tissues exists diversity.Senescence-accelerated mouse of oral-breathing model suffers more severely in tissue impaired in upper airway muscle and mucosa,musculus rectus abdominis and leukocytes.Normal accelerated mouse of oral-breathing model suffers more severely only in upper airway muscle.
