Objective:Vx-2 tumor cell lines was widely used in the establishment of clinical study of various diseases. It is an easily transplanted tumor, growing faster and has invasion which are the characteristics of it's morphological and biological. The tumor was tested for squamous cell carcinoma and were similar to vaccination Rabbit in various types of multiple sites. We use rabbit Vx-2 tumor cell lines established oral Vx-2 tumor animal models, expecting for providing an applicable animal model for further research on oral and maxillofacial tumors.Method:Firstly, We made Vx-2 tumor cell lines recovered for getting the transplant tumor tissue to establish rabbit model with the tumor. Thirty New Zealand white rabbits with 1.5 ~ 2.0 kg weight were randomly divided into two groups. They were kept in standard laboratory to ensure that the media has high similarity. In our study we used two ways in the mouth and extra the mouth to establish the rabbit cheek Vx-2 tumor model and then through observation of rabbits in the spirit , appetite and activities to compare the two groups. By measurement of the tumor size we had more date. The HE staining and flow cytometry were tested to compare whether the two groups had significant difference in tumor properties and have a preliminary exploration of the rabbit Cheek Vx-2 tumors.Results:In our experiment the two ways of transplantation can establish the successful rabbit cheek Vx-2 tumor model. The whole process was more simple and had higher success rate. There were no signification difference of the two groups in the spirit ,food and drink which are the basic characteristics of life. From the Observation of animal model we found that the Vx-2 tumors underwent the stages of rapid tumor growth, necrosis in tumor Centre and surface rupture, with about 4 to 6 weeks life. We also found that in the tumor size and HE staining the two groups had no significant difference. It was the same as many scholars. The tumor apoptosis rate compared between two groups show no significant difference (P> 0.05).Conclusion:The rabbit oral squamous cell carcinoma mode by Vx-2 tumor tissue transplantation was more easy and had shorter time. It had stable nature, reproducible, high rate of tumor formation, simple operation and performed squamous cell carcinoma in histological observation. All of these were show that this method will provide a larger and better animal model for the study of OSCC. Our study also tell us that the two ways of implantation had higher similar in the nature of tumor and tumor growth .
