Galangal,with warm stomach for dispelling cold,digestion and pain relief,which is the dried roots of ginger plant.Galangal can be divided into two different kinds: the small ginger is native to Chinese herb,which has the cone inflorescence of white roses;the other is larger ginger,perennial herb grown in southeastern Asia.Galangal is rich in flavonoids and phenolic acids such as phenolic compounds,which has also been used as spices in the traditional food.In recent years,the biological activity of galangal has been studied in depth,it is reported that it has anticancer,antibacterial,antiulcer,antifungal,insecticidal and other characteristics.In many countries,galangal has become cheaper and more accessible medicinal herbs,and has been widely used in flood,analgesia,anti-inflammatory,antimicrobial and other medical purposes.Galangin,as the main active substance in the extraction of galangal,which has many pharmacological activities.In this experiment,shows that the alcohol extraction of galangal and galangin have the function of reducing uric acid,anti-gouty arthritis.Due to its poor solubility in vitro and the low bioavailability in vivo,the application of clinical treatment is limited.this subject do research on the granular as an agent to increases the solubility of galangin in vivo,so the full text is divided into the following five parts:Chapter one: ReviewsThis part mainly focuses on the physicochemical properties,pharmacological activities,research progress of galangal preparations and the applications of granules.Establishing the theoretical foundation for the study of pharmacokinetics,pharmacodynamics and galangin preparation.Chapter two: Research on the extracting technology of galangalAccording to the literature,the galangin is the main active ingredient of galangal,so this chapter first carries on the water extract and the alcohol to the Gao Liangjiang,Therefore,the test wavelength of galangin was detected by ultraviolet and to establish the HPLC method for determining the content of galangin in vitro.The alcohol extraction of galangal was proved to be the effective part.At the same time,using one-factor experimental design and orthogonal experimental design to select the optimum extracting method of galangin.Chapter three: Separation and purification of galangalIn this chapter,to screen the effective method of extracting galangal,using various macroporous adsorption resins to test the adsorption and desorption capacity of galangal.To establish a simple,fast and efficient method of separating and purifying the galangin from the galangal alcohol extraction.Chapter four: Study on the extaction of galangal reducing uric acidIn this chapter,the HPLC analysis method of galangin and the uric acid model were established.Collecting blood time was determined by the high concentration of uric acid in the 3h after making uric acid model,which was proved that the galangal alcohol extraction is the mainly active part.Screening the the high,medium and low doses of galangal alcohol extraction.Proving the galangin was the main active part in galangal alcohol extraction to reduce uric acid in the gout.Chapter five: Study on the anti-gouty arthritis of galangalIn this chapter,by injecting a certain dose of uric acid sodium in the ankle joint of the rat foot,to cause swelling of the joints and establish model of gouty arthritis.Measuring the circumference of the ankle joints of rats after the injection of monosodium urate by Vernier caliper and calculating the swelling degree of the ankle joint.Taking the rat orbital serum to test the content of IL-1?,IL-6,TNF-? and COX-2 inflammatory factors in the serum of rats,which was proved that galangin is the active part of galangal against gout.Chapter six: Prepation and evaluation of galangal granules in vitro and in vivoIn this chapter,the galangal alcohol extraction was mixed with the auxiliarymaterial in a certain proportion by the wet-granulation to prepare galangal granules,and the optimum prescription of galangal granules was screened by orthogonal design.Finally,the quality of galangal granules was evaluated in vitro from the aspects of roundness,yield,moisture absorption and loading differences and the effect of reducing uric acid was also studied in vivo.
