The Pharmacy And The Main Pharmacodynamics Studies Of Tonglishu Granules

Objective:The prescription of Tonglishu Granules has significant clinical efficacy.The research group was studied in terms of preparation process and quality standards,and the main pharmacodynamics test for gout treatment,providing experimental basis for the development of the preparation and clinical treatment of gout.Method:Through orthogonal test design and other methods,the best extraction process for each part of the prescription drug was optimized and determined.Through the multi-index comprehensive scoring method,the molding process of the prescription was optimized.Qualitative identification for Gardenia jasminoides Ellis,Pueraria lobata?Willd.?Ohwi,Salvia miltiorrhiza Bge.,Paeonia lactiflora Pall,Glycyrrhiza inflata Bat.in the prescription were studied by thin-layer chromatography,the research group determined the content of Puerarin by High Performance Liquid Chromatography,and formulated the quality standard of Tonglishu Granules.The efficacy and safety of the preparation were evaluated through experimental study on the pharmacodynamics of animals.Results:Volatile oil was extracted from Atractylodes macrocephala Koidz.and Atractylodes chinensis?DC.?Koidz.in Tonglishu Granules.Pueraria lobata?Willd.?Ohwi,Salvia miltiorrhiza Bge.were extracted by 65%ethanol.The water extraction and alcohol precipitation process was adopted to the residue and the remaining 11 herbs,and the respective optimum process parameters were determined.The inclusion of volatile oil was mixed with soluble starch and extract,and 94%ethanol was used as a wetting agent to prepare and form.The five herbs of Gardenia jasminoides Ellis,Pueraria lobata?Willd.?Ohwi,Salvia miltiorrhiza Bge.,Paeonia lactiflora Pall,Glycyrrhiza inflata Bat.in the granules were identified.The results showed that the spots were round and clear,the degree of separation was good,and the negative was no interference;The content of Puerarin in the granules was determined.The content of Puerarin(C₂₁H₂₀O₉)per bag was not less than 15.70mg.Tonglishu Granules have anti-inflammatory,analgesic,and relief of gouty arthritis ankle swelling degree,lowering blood uric acid pharmacodynamic effects.In the acute toxicity test,mice were administered 150.6 g/kg granules cumulatively,which was 292.83 times that of clinical adults.No toxicity was observed in mice.Conclusion:Through the research on the preparation process,quality standard and main pharmacodynamics of Tonglishu Granules,it was shown that the preparation process of Tonglishu Granules was scientific and reasonable,the quality controllability was strong,and it was safe and effective.
