Objective: To research the qualitative and relative quantitative expression of Connexin43 in maternal membranes and placenta with oligohydramnios and normal,To analyze its important role in the occurrence of oligohydramnios and its possible regulatory mechanism and effect.Methods:Choosing between April 2014 and December 2015 in Lanzhou university first hospital of obstetrics and gynecology by cesarean section of 57 cases of term maternal as the research object,including the 27 oligohydramnios maternal cases as test group,30 normal amniotic fluid cases as contrast group.Using SABC immunohistochemistry and RT-PCR technique to disclose the expression and localization of Cx43 protein and m RNA in the two groups of maternal placenta and fetal membranes.Results:1.The expression of Cx43 protein was detected by SABC immunohistochemis try in two groups of membranes and placenta.2.The positive rate of Cx43 protein expres sion of oligohydramnios group in the epithelial cells of the membranes was significantly d ecline than those in contrast group,the difference was markedly significant(c2=5.747,P=0.017),the positive rate of Cx43 protein expression in placental trophoblast was obviously below the contrast group,too.the difference was statistically significant(c2=3.986,P=0.046).3.Cx43 m RNA was detected by RT-PCR in both fetal membranes and placenta.4.The expr ession of Cx43 m RNA in the oligohydramnios group was 0.51 times of that in the contra st group,markedly below the contrast group,no obvious difference(t=15.792,P<0.01),a nd in the placenta was 0.54 times of that in the normal group,also distinctly decline than the contrast group,no obvious difference(t=14.782,P<0.01).Conclusion: 1.Cx43 protein and m RNA in membranes,human placenta are expressed,mainly in amniotic epithelial cells,and chorionic trophoblast cells and placenta trophoblast cells.2.The expression of Cx43 protein and m RNA in fetal membranes and placental tissues of patients with oligohydramnios were down-regulated.3.Cx43 perhaps makes a major effect in the amniotic fluid absorption pathway and maternal fluid exchange in the maternal amniotic fluid.The expression of Cx43 may be involved in the amniotic fluid abnormalities.The mechanism of expression in the membranes is different from that in the placenta.In addition to the water permeation function,alsowith apoptosis,and the specific function needs further study.4.The Apgar score of neonates with oligohydramnios decreased compared with Apgar score of normal term neonates,Compared with significant differences.It is speculated that oligohydramnios can affect the outcome of pregnancy and increase fetal hypoxia and neonatal asphyxia incidence.
