Objective:To discuss the differences and the relationship between the expressions of AQP9andCaspase7in placenta and membranes of oligohydramnios and normal group.Methods:We Choose30cases of oligohydramnios and30cases of normal maternal whodelivere in37-41weeks by elective Caesarean section in June2013to November2013inobstetrical department of affiliated hospital of Yan’an university. Age, pregnant weeksand the number of pregnancy of two groups had no significant difference.The two groupshave not a recent history of medication, infection, and maternal dehydration and soon,both eliminating medical and surgery complication and the other obstetricalcomplication (gestational hypertension,premature rupture of membrane,fetal growthrestriction,congenital malformation and so on).Immunohistochemistry was performed todetermine the area and level of expressions of AQP9and Caspase7in placenta andmembranes from a test group (30cases of oligohydramnios) and a control group (30cases of normal pregnancy).And we detect the expression intensity of the two kinds ofprotein by semi-quantitative analysis.Results:1.The expression of Caspase7and AQP9in the placenta of two groupsIn the cell of placenta of two groups maternal,Caspase7and AQP9protein arepositive expression and appear brown-yellow.The areas of positive expression are mainlydistributed in the cell membrane and cytoplasm of placenta. Immunohistochemistry showthe expression of caspase7in the cell membrane and cytoplasm of placenta is higher in the test group than the normal group(t=2.85),and the expression of AQP9of them islower in the test group than in the normal group(t=2.32). There are significant differencesbetween the two groups,(P<0.05).2.The expression of Caspase7and AQP9in the membranes of two groupsIn the cell of membranes of two groups maternal,Caspase7and AQP9protein arepositive expression and appear brown-yellow.The areas of positive expression are mainlydistributed in the epithelial cells of membranes.Immunohistochemistry show theexpression of caspase7in the epithelial cells of membranes is higher in the test groupthan the normal group(t=3.12),and the expression of AQP9of them is lower in the testgroup than in the normal group(t=4.16). There are significant differences between thetwo groups(P<0.05).3.The relation of Caspase7and AQP9expression in the the placenta and membranesof two groupsThe Spearman grading correlation analysis showed the correlations betweenCaspase7and AQP9of maternal placenta and membranes in normal group respectivelyare0.12and0.23(rs=0.12, P>0.05;rs=0.23,P>0.05).There were no significantdifferences.And the correlations of them in test group respectively are-0.63and-0.78(rs=―0.63,P<0.05;rs=―0.78,P<0.05). There are significant differences between thetwo groups(P<0.05).Conclusion:1.There are significant differences in the expression of Caspase7and AQP9in thecell of membranes and placenta between the two groups. So we guess higher expressionsof caspase7and lower expressions of AQP9in placenta and membranes may playimportant roles in oligohydramnios.2. In placenta and membranes,The expression of caspase7was not significantlyrelated to that of AQP9in normal group.However,the expression of caspase7in placentaand membranes was inversely correlated with that of AQP9in oligohydramnios. So weguess higher expressions of caspase7in placenta and membranes of oligohydramnios can induce cell apoptosis, reduce the expressions of AQP9in placenta and membranes andimpact the structure and function of AQP9.The change of AQP9lead to materia exchangeimpairment, water metabolism impairment, reduce water exchange between maternal andfetusin and cause oligohydramnios.
