Objective:To explore the expression and localization of AQP11 in human term pregnancies with oligihydramnios and polyhydrammnios, and it’s role in amniotic fluid balance.Methods:82 patients under went elective cesarean sections were studied,which came from the First Hospital of LanZhou University from April in 2013 to July in 2014.25 patients with isolated oligohydramnios,27 with isolated polyhydramnios and the others 30 with normal amniotic fluid volume. Immunohistochemistry and reverse transcription polymerase chain reaction were employed to determine expression and localization of aquaporin 11 in amnion, chorion and placenta.Results:1.The expression of AQP11 protein was detected in amnion, chorion,trophoblast and placenta using IHP.2.AQP11 protein expression was decreased in amnion of oligihydramnios, increased in chorion and placenta of oligihydramnios compared with normal AFV group,the differences were obvious;AQP11 protein expression was increased in amnion and chorion of polyhydramnios, decreased in placenta of polyhydramnios compared with normal AFV group,the differences were obvious;3.The expression of AQP11 mRNA was detected in amnion, chorion and placenta using RT-PCR.4.AQP11 mRNA expression was 0.59 times in amnion of oligihydramnios 1.71 times in amnion and 2.53 times in chorion of polyhydramnios,the differences were obvious;AQP11 mRNA expression was 0.71 times in placenta of oligihydramnios,the difference was statistically significant.Conclusions:1.AQP 11 protein and mRNA are expressed in amnion,chorion andplacenta,located in epithelia of amnion, trophoblast of chorion,trophoblast and vascular endothelial of placenta.2.The changes of AQP11 in amnion,chorion and placenta of oligihydramnios and polyhydrammons may be involved in the regulation of abnormal AFV,the changes of AQP11 expression in amnion are the main causes of abnormal AFV,the changes of AQP11 expression in placenta may be the results of regulating in order to adapt the changes of AFV.3.The expression and function of AQP11 in chorion is different from amnion andplacenta,in addition to the water permeability functions there may have other potential functions, and its potential functions should be further studied.
