Up-regulation Of LncRNA AC006050.3 In Gastric Cancer And Its Clinical Association

BACKGROUND: With the progress of medical and health technology around the world,cancer will be the leading cause of morbidity and mortality in most parts of the world in coming decades.Gastric cancer as one of the world's highest incidence and mortality of the tumor,the pathogenesis and early detection of cancer has been the focus of research to overcome gastric cancer.Long non-coding RNA(lncRNA),with size larger than 200 nucleotides,is a new class of the non-protein coding RNA.In recent years,as a new field of tumor molecular mechanism research,lncRNA has been found to be involved in the occurrence and development of many tumors and has played a crucial roles in many regulatory progress.Lnc RNA because of its extensive interaction with DNA,RNA,protein and stronger tissue specificity,its participation in gene regulation mechanism is more diverse.OBJECTIVE: To explore the expression of lncRNA AC006050.3 in gastric cancer and its relationship with clinical and pathological features.METHOD: Acordding to the gene chip results of fourteen early gastric cancer,the expresstion of lncRNA AC006050.3 in tumor is 8.261 times higher than normal tissue(P<0.01).By reviewing relative literature,regard lncRNA AC006050.3 as the target research gene.A total of 42 samples of gastric cancer patients were collected by surgical procedure,and the corresponding c DNA samples were obtained by total RNA extraction and reverse transcription PCR.The expression level of lncRNA AC006050.3 in gastric cancer tissues and non-tumor tissues was detected by real-time fluorescence quantitative PCR(RT-PCR).And further verified by agarose gel electrophoresis.The data were verified by statistical methods such as chi-square test and Fisher's exact test.RESULT: The expression level of lncRNA AC006050.3 was correlated with the diameter of tumor(p = 0.018),which was statistically significant.In addition,the expression level of lncRNA AC006050.3 has no statistically significant difference with other clinical and pathological features,such as gender(p = 0.699),age(p = 0.464),degree of differentiation(p = 0.379),infiltration(p =0.699),lymph node metastasis(p = 0.715),TMN staging(p = 0.222).The area under the curve of ROC curve of gastric cancer tissue of lncRNA AC006050.3 was 0.748,the best cutoff value was0.0153(sensitivity 100%,specificity 48.93%).CONCLUSION: lncRNA AC006050.3 is up-regulation expressed in gastric cancertissues.Besides,The expression level of lncRNA AC006050.3 was correlated with the diameter of tumor(p = 0.018),which was statistically significant.
