Objective: To observe the effect of Dachengqi Decoction " timely purgation to preserve yin " on endotoxemia rat serum endotoxin,cAMP and cGMP expression in colon,common probiotic lactobacilli and endotoxemia conditional pathogenic Escherichia coli proliferation,and explore its treatment of endotoxemia " timely purgation to preserve yin " mechanism,providing modern medical explanation for rich Chinese characteristics clinical use of Dachengqi Decoction in the treatment of endotoxemia,provide new ideas and methods to study the theory of prescription compatibility.Method: The first part,Dachengqi Decoction acute Yin effect on serum endotoxin and colon cAMP,cGMP in endotoxemia rats.SD rats were randomly divided into control group,model group,Dachengqi Decoction high dose group(5g/kg · q12h),middle dose group(2.5g/kg · q12h),low dose group(1.25g/kg · q12h).Control group underwent sham operation,while other groups of endotoxemia model induced by cecal ligation and puncture operation,administration of 5 times,sampling detection of serum endotoxin and colon cAMP,cGMP level.And take pictures with Leica microscope,processing by the image-pro-plus6.0 gets the average optical density value.The second part,the effect of Dachengqi Decoction " timely purgation to preserve yin " on common probiotic Lactobacillus and endotoxin conditioned pathogen Escherichia coli proliferation.Using spectrophotometer to detect Dachengqi Decoction(10-1~10-7)on the in vitro effect of Escherichia coli and Lactobacillus proliferation.Real-Time PCR detection of Dachengqi Decoction high dose group(5g/kg · q12h),middle dose group(2.5g/kg · q12h),low dose group(1.25g/kg · q12h).have the effects of in vivo on endotoxemia rats of Escherichia coli and Lactobacillus proliferation before modeling and after modeling 8h(before the first dosage),2d,7d.Results: The first part,Serum endotoxin,c AMP,c AMP/cGMP of Dachengqi Decoction high and middle dose group were significantly decreased,compared with the model group,there was significant difference between the two groups(P < 0.01);The levels of endotoxin in low dose group decreased,compared with the model group,there was significant difference(P < 0.01);Low dose group cAMP/cGMP ratio decreased,compared with the model group,the difference was statistically significant(P < 0.05);cGMP of high dose group increased significantly,compared with the model group,there was significant difference between them(P < 0.01);The middle dose group increased cGMP,compared with the model group,there was statistically significant(P < 0.05);On the decrease of serum endotoxin,cAMP,cAMP/cGMP ratio,increasing the level of cGMP,the high dose group and middle dose group is better than the low dose group(P < 0.05).The second part,In vitro experiment:Dachengqi Decoction(10-3~10-6)has certain promoting effect on the growth of lactobacilli;10-1?10-2?10-7 on the growth of Lactobacillus have certain inhibition;10-1~10-7 has certain promoting effect on the growth of Escherichia coli.In vivo experiment:(1)After modeling 8h(before the first dosage),high,low dose group of Escherichia coli increased significantly;After modeling 2d,high,middle dose group of Escherichia coli was significantly reduced,the rapid increase in the number of lactobacilli;Model 7d,the Escherichia coli recovered to normal levels,high,middle dose group of lactic acid bacteria is slightly higher than normal level;(2)Low dose of Dachengqi Decoction diarrhoea dispel bacteria effect is weak,almost no role in promoting the growth of lactobacilli,after making the mould number 7d of Lactobacillus decreased;(3)High,middle dose of Dachengqi decoction had the enhancement of lactic acid bacteria: Escherichia coli ratio,low dose is almost no effect.Conclusion:Dachengqi Decoction could treat endotoxemia,its " timely purgation " mechanism may be related to the decrease of serum endotoxin level and dispel the conditional pathogenic Escherichia coli,the " preserve yin " mechanism may be related to the decrease of cGMP,reduce the ratio of cAMP/cGMP and promote the proliferation of probiotic lactobacillus.
