| Endotoxemia is the basic pathology of various infective illnesses. To effectively control the developments of endotoxemia is very important to reduce the death rates made by various infective illnesses. Dachengqi decoction is one of the Ancient prescriptions of traditional Chinese Medicine. In clinical it is a good cure for endotoxemia which is coursed by various infective illnesses.But unfortunately we haven’t fully understand the mechanism of action or pesticide effect of material basis, so to some degrees it limited the use in clinical and new medicinal research of this prescription.Objective This text is about an research in rats who have been made models by the method of abdominal cavity infection via intraperitoneal injection of rat feces suspension,in order to induce endotoxmia. After the rats are made models, we watch the cure action of enterogenous endotoxemia of the rat under the prescriptions of Dachengqi decoction and most part of Into-blood-monomers such as rhein, gallic acid, naringenin, magnolol, honokiol. And do some research about the mechanism of action,so we could have some further understand about their material basis.Method90SD rats were divided into10groups randomly: control group, model group, Dachengqi decoction group, Rhein group, Gallic Acid group, Naringenin group, Magnolol group, Honokiol group and Monomer Combination group. Every group there are10rats, half male and half female.Rats were orally administrated2days before modeling.At the third day the rats of model group, Dachengqi decoction group, Rhein group and Gallic Acid group were made models by the method of abdominal cavity infection via intraperitoneal injection of rat feces suspension,in order to induce endotoxmia.and then rats would be administrated1more day. We need to observe the signs and symptoms of the rats,together with the Composition in the form of livers, lungs and kidney. Then quantificationally measure the number of endotoxemia in the rats’blood plasma and the number of three Inflammatory cytokines such as IL-10、IL-1β、TNF-a in the rats’blood serum.So we can do some research about the cure actions of Dachengqi decoctions to endotoxemia, and find mechanism of action and material basis.Result Compared with control group, endotoxemia in blood plasma of model group and each curing group rose a lot(P<0.01).And the signs and symptoms of the rats were unusual, histomorphology research find the pathological changes of inflammation.It means that the models of enterogenous endotoxemia are well done. Compared with model group, enterogenous endotoxemia in each curing group markedly reduced (P<0.01),and histomorphology research find that the pathological damages have been better, it means that the medicine used in each curing group have obviously treat the rats. And compared with Dachengqi Decoction group, enterogenous endotoxemia in monomer matter curing group(Rhein group, Gallic Acid group, Naringenin group, Magnolol group, Honokiol group) are in a high level,it means that Dachengqi decoction has better effect in cure of enterogenous endotoxemia than monomer matter.Compared with the control group, the number of blood serumIL-10、IL-1βand TNF-a of the rats in model group and each curing group were obviously promoted (P<0.01),it means that endotoxemia lead to the inflammatory reaction of organism; Compared with model group, the number of blood serumIL-10、IL-1βand TNF-a of the rats in each curing group were evidently cutting down (P<0.01),it means that Dachengqi decoction and the above monomer matter can reduce the number of inflammatory cell of rats in different degrees.Conclusion Dachengqi decoction and its main Into-blood-monomer such as rhein, gallic acid, naringenin, magnolol, honokiol,they effectively treat the rat who have got the illnesses of the experimental Enterogenous Endotoxemia. And they are the antagonism of the endotoxin in the rat. In the same time they could reduce the damages to the internal organs of the body.The mechanism of action is ralated to the restriction of the level of the Inflammatory cytokines such as IL-10、IL-1β and TNF-a, It also has something to do with adjusting the balance between pro-inflammatory mediator and Inflammatory medium. And rhein, gallic acid, naringenin, magnolol, honokiol are the main material basis of Dachengqi decoction to cure the Enterogenous Endotoxemia... |
