| Objective: To develop the animal model of Syndrome Suitable for Dachengqi Decoction in rabbits, to establish stability modeling method, and to explore the further function of purgative treatment.Methods: The 60 experimental rabbits were divided into four groups randomly and equally:Control group,Model group,Model-treating group and Model-proving group. Animals of Model group were given The Bismuth Subcarbonate feed for 2 days,and no water in the second day, then injected E.coli Endotoxin by the ear vein; Dachengqi Decoction was administered through pouring into stomach to Model-treating group before 1h and after 2h the injection of Endotoxin, and Dachengqi Decoction was given simultaneously when Model-proving group was given the Bismuth Subcarbonate and Endotoxin. At the same time, Normal Saline was injected in Control group for demands. After six hours of infection, blood was taken from the heart for indicators testing. Finally, we killed all animals by air, and observed the pathological changes of lung,liver,intestine and kidney under the microscope. Throughout the whole experimental process, measured the temperature and abdominal circumference regularly, and recorded the general situation objectively and accurately.Results: Control group had no obvious abnormality. The temperature and abdominal circumference of Model group animals increased significantly. WBC counts and NO,MDA,MMS content increased obviously, while SOD decreased; K and Na content decreased in varying degrees; Whole blood viscosity and plasma viscosity increased, whereas erythrocyte deformation index decreased; The content of t-PA decreased, while PAI-1 content increased to some extent. Furthermore, TXB2 and TM value also increased. Compared with control group, there are some significant differences in Model group (p<0.01 or p<0.05), while no or partial indicators have some significant differences in Model-treating group and Model-proving group. Pathological observation: Most model group animals had macular bleeding spots under the skin, obvious congestion or hemorrhage in most of organs and tissues, while tumefaction in intestines. Microscopic observation: in Model group, lung consolidation, pulmonary sepatal thickening was turned out; Intestinal mucosa and liver had cellular degeneration and necrosis, congestion and hemorrhage of tissues, infiltration of inflammatory cells, but kidney changed scarcely. At the mean while, only minor pathological changes were found in Model-treating group and Model-proving group. Conclusions: 1. Through the Bismuth Subcarbonate feeding, water cuting and Endotoxin infecting ,the main sumptoms,signs and pathological changes of Model group animals have very close interrelationship with Syndrome Suitable for Dachengqi Decoction in Treatise on Exogenous Febrile Disease, so these modeling methods can be successful. 2.The Dachengqi Decoction can not only reduce the temperature and the abdominal circumference obviously, alleviates the symptoms and signs of the rabbits effectively, but also improve various indicators and pathological changes. So these results have proved the existence of the model of Syndrome Suitable for Dachengqi Decoction, and also indicate the reliability of the modeling method.. |
