Objective:Chemotherapy is one of the most important methods for the treatment of malignant tumors.The chemotherapy drugs can bring many adverse effects to the body,including the digestive system toxicity,that can cause great pain to patients.Chemotherapy induced gastrointestinal inflammation injury,limiting the long-term use of large doses of chemotherapy drugs,but also reduce the compliance of patients,affecting the efficacy of chemotherapy.This paper intends to investigate the preventive effect of Xiaobanxia Decoction(XBXT)on gastrointestinal inflammation injury in rats after chemotherapy,and preliminary study of its mechanism,to provide experimental basis for clinical treatment of Gastrointestinal tract inflammation injury(GIM)induced by cisplatin with small Banxia Decoction(XBXT).Mothods:The chemotherapy rat model was established by intraperitoneal injection of6mg·kg?1cisplatin.The Wistar rats were randomly divided into normal group,XBXT normal control group,cisplatin model group,positive drug omeprazole group,XBXT low,middle,high dose groups.The rats in omeprazole group,XBXT normal control group,XBXT low,middle and high dose groups were respectively given 2.4 mg·kg?1·d omeprazole,0.8,1.6 and 3.2 g·kg?1·d XBXT by oral gavage before modeling 2 d and then twice a day.The rats in normal control group and model group were gavaged with equal volume of distilled water.After modeling 24 h and 72 h,TNF-?,IL-1? and PGE2 were measured by Elisa.The positive cells of NF-?B,COX-2 in rat sinuses ventriculi and ileum were detected by immunohistochemistry method.Sinuses ventriculi and ileum were visualized by hematoxylin and eosin staining.Results:Model group of rats food,water and body weight were significantly reduced,appear thin bowel,listlessness,irritability and other phenomena,each treatment groups was significantly improved include food,water,body weight and general behaviorin rats of 72 h.Inflammatory pathological changes were observed in the gastrointestinal mucosa of rats in model group,levels of TNF-?,IL-1?,PGE2 increased significantly in serum(P<0.05),NF-?B,COX-2 expression levels weresignificantlyhigher than normal group in Sinuses ventriculi and ileum.XBXT can improve chemotherapy rats gastrointestinal inflammation,the middle and high dosages of XBXT could significantly reduce the levels of TNF-?,PGE2 at 24 h and 72 h in cisplatin-treated serum.IL-1?decreased significantly at 72 h in cisplatin-treated serum(P<0.05),and can significantly reduce the positive expression of NF-?B,COX-2 in gastric and ileum tissue at 72 h.Conclusion: XBXT can prevent and treat the gastrointestinal inflammatory injury,induced by cisplatin in rats,and its mechanism may be related to the inhibition of inflammatory factor expression.
