Objective:The relationship between HOXA1 gene and endometriosis with infertility was investigated by detecting the expression of HOXA10 gene in the endometrium of EMT infertile patients at the transcriptional level and translation level.Method:In this study,we selected 23 infertility patients with EMT who were included in the gynecological inpatient department of the First Affiliated Hospital of Kunming Medical University from January 2015 to September 2016,25 cases of non-EMT patients who met the inclusion and exclusion criteria.According toASRM Modified stage of endometriosis staging(1997)staging,I 2 cases,II 9 cases,III 8 cases,IV 4 cases,18 cases of primary surgery,3 cases of the second operation,2 cases for the third operation.All patients included in the record after admission,menstrual cycle,drawn time,endometrial thickness,E2,P and other general information,EMT infertility group need to record infertility years,the history of previous treatment.In the menstrual cycle on the first 20-24 days to take its own endometrium.The relative expression of HOXA10 mRNA and protein was detected by real-time PCR and Western blot.The results were statistically analyzed by SPSS 17.0,p<0.05 was statistically significant.Results:1.There was no significant difference in age,menstrual cycle,time of extraction,endometrial thickness,E2 and P between EMT patients and non-EMT patients(p>0.05).The mRNA expression of HOXA10 was 1.52 ± 1.07 and 1.34±0.97,respectively.There was no significant difference between the two groups(P =0.496).At the level of translation,the expression of HOXA10 mRNA in EMT infertile patients and non-EMT patients was 1.52 ± 1.07 and 1.34 土 0.97,The expression of HOXA10 protein in EMT infertile patients and non-EMT patients was 0.83 ±0.28,1.05 ± 0.32,respectively.There was a significant difference between the two groups(p = 0.02),EMT infertility group was lower than that of non-EMT EMT group.2.In the patients with stage I-II and III-IV EMT,the age and infertility age of group III-IV EMT were higher than those of stage I-II in patients with different degree of EMT infertility,the difference was statistically significant(P = 0.027,p = 0.026),menstrual cycle,time of extraction,endometrial thickness,E2 and P were not significantly different(p>0.05).The expression levels of HOXA10 mRNA in the endometrium of stageⅠ-Ⅱ EMT group and stage III-IV EMT group were 1.24 ±0.74,1.25± 1.02,respectively.There was no significant difference between the two groups(p The expression levels of HOXA10 protein in EMT group and stage III-IV EMT group were 0.94 ± 0.23 and 1.07 ± 0.24,respectively.The expression of HOXA10 protein in the endometrium was 0.94 ± 0.23 and 1.07 ± 0.24,respectively.Differences(p = 0.413).Conclusion:1.The down-regulation of HOXA10 gene expression in the endometr,ium of EMT patients may be related to the pathogenesis of infertility.2.HOXA10 gene is a sensitive molecular biology indicator to evaluate the endometrial receptivity of EMT infertile patients. |
