Objective By comparing HOXA10(Home box, HOX) protein and mRNA expression in the eutopicendometrial tissue between normal women and women with endometriosis, in order to discuss theHOXA10gene on the pathogenesis of endometriosis associated infertility.Methods We choose Patients that in treatment in the affiliated Hospital of Ningbo University fromOctober2011to September2012that initial diagnosed as endometriosis and uterine leiomyoma, preparedto surgical treatment and have regular menstrual cycle. Get a small amount of endometrial tissue during thesurgical operation. Postoperative pathology confirmed cases of preliminary diagnosis and in accordancewith the inclusive criteria as the research object. Selected cases is divided into two groups according topathological diagnosis, each group have thirty cases. Patients who are diagnosed as adenomyosis, ovarianendometrial cyst and endometriosis of pelvic peritoneum were in endometriosis group (E group). Patientsdoesn’t have complications and met the diagnostic criteria were in control group (N group). Endometrialtissue is undergo HE (Hematoxylin-Eosin) staining in order to determine the histological staging,immunohistochemical staining method and real-time PCR were used to detect the expression of HOXA10protein and mRNA respectively.Results In group E, we find out the endometrial tissue of11cases in the proliferating phase (P phase),the rest19cases in secretion phase (S phase) including early secretion phase (S1phase)12cases andmid-late phase (S2phase)18cases. In group N,10cases in P phase and the remaining20cases in S phaseincluding S1phase9cases and S2phase11cases. HOXA10protein express in both endometrial epithelialand stromal cells in those two groups. In group N, compare to P phase, the expression of HOXA10proteinand mRNA is markedly increase in S2phase (P<0.01); In group E, we doesn’t see similar obviousincreased in the expression of HOXA10protein and mRNA in S2phase compared with P phase. Comparedthe two group of S2phase, the expression of HOXA10protein and mRNA in group N is significantlyhigher than group E (P<0.01).Conclusion HOXA10gene express in both endometrial epithelial and stromal cells of the eutopicendometrial tissue during the menstrual cycle of the normal women and women with endometriosis, thereis a higher expression in endometrial tissue at mid-late secretion phase than proliferating phase and earlysecretion phase in normal women. However, no significant increase was found at mid-late secretion phasein women with endometriosis. HOXA10gene expression peak does not appeared prompt lower expressionof HOXA10gene in the mid-late secretion phase and implantation period result in the damage of theendometrial receptivity and maybe this is one of the possible mechanisms of EM related infertility.
