Backfound Psoriasis is a chronic inflammatory skin disease,The etiology is not clear,no effective method to cure,it bring double injury to body and mind of psoriatic patients and families.The pathogenesis of psoriasis is closely related to the T cell immune disorders,Th17/Treg cells play an important role in the process of psoriasis occurs.Cytokines that MSCs secrete have immune regulation function for T cells.MSC can inhibit the shortness of inflammatory activation of T lymphocytes,value-added and matured and change the immune cell secretion of cytokines,anti-inflammatory and immune tolerance to make it tend to functior,and it also can promote inhibiting inflammatory response of T lymphocyte differentiation advantage,in order to help the body to restore immune balance.Stem cells possess the property of the kidney essence,and it have similar with the kidney essence on sources,distribution and function of "kidney essence" in addition,Bushenpeiyuan medicine have functions of regulating biological characteristics of MSCs.Base on the previous study and the experience of prestigiouschinese physician,professor Song applied a new method"KaiTongXuanFu,BuShengPeiYuan" to treat psoriasis patients,and receive good effect.The main content of this paper is,For the first time,base on previous work,and put forward the research hypothesis about the effect mechanism:the pathogenesis of psoriasis is closely related to the T cell immune disorders.As the upper part of T cell immune network imbalance,It is a key to lead to the pathogenesis of psoriasis for abnormal immunity function of MSC inpatients with psoriasis."KaiTongXuanFu,BuShengPeiYuan" method can adjust the peripheral blood of Th17 and Treg cells and secretion of cytokines through the intervention function of MSC.We researched immune regulation function of MSC for T cells by modern technologies to test the hypothesisObjective 1.To study the differences of the biological characteristics and immune regulation function of adipose mesenchymal stem cells(AMSCs)on psoriasis vulgaris patients and healthy individuals.2.To investigate the immune regulation function of adipose mesenchymal stem cells on peripheral blood mononuclear cell in psoriasis vulgris patient(PBMC).3.To investigate the impace of medicinal indiamulberry riit poiysaccharide(MOP)inimmune regulation function of adipose mesenchymalstem cells(AMSCs)on psoriasis vulgar patients.Methods 1.AMSCs were isolated and cultured from human psoriatic and healthy adipose tissue,the phenotypes and cell cycle of AMSCs taken from three generation were detected by flow eytometry.Alkaline phosphate enzyme staining and oil red o staining were used respectively to identify their adipogenic and osteogenic capacity.Next,the levels of inflammation antimicrobial pro inflammatory factor were detected by PCR and ELISA.2.PBMC in psoriasis vulgaris were co-cultured with AMSCs on psoriasis vulgaris patients and healthy individuals.Thl7and Treg proportion were analyzed by flow cytometry,the levels of inflammation antimicrobial pro inflammatory factors in PBMC were detected by PCR and ELISA.3.Medicinal indianmulberry root polysaccharide(MOP)with and without pretreated AMSCs on psoriasis vulgaris patients were used as MOP-pretreated group and MOPunpretreated group.The biological characteristics and immune regulation function of MOP-pretreated group and MOP-upretreated group were analyzed based on flow cytometry,alkaline phosphate enzyme staining,oil red O staining and PCR.Next,they were co-cultured with PBMC in psoriasis vulgaris respectively,the levels of inflammation antimicrobial pro inflammatory factors in PBMC were detected by PCR.Results We found that there were no significant change in cell morphology,and cell surface markers were expressed high for CD29?CD44?CD73,while lower for CD31?CD45 and HLA-DR.AMSCs of psoriasis patients and healthy people both had the ability of adipogenic and osteogenic differentiation.But the cell cycle showed the third generation AMSCs proliferation rates were slower than of normal control,as compared with healthy controls,adipogenic differentiation ability was stronger.What' more,the levels of inflammatory cytokines in psoriasis group was lower than that in controls such asIL-10?IDO?TGF-?,on the contrary the levels of pro inflammatory factor in psoriasis group were higher than that in controls,such as TNF-a,IFN-Y.Next,We found that AMSCs on psoriasis vulgaris patients and healthy individuals increased the Treg proportion of total lymphocytes.And increased the levels of FOXP3?IL-10?TGF-?1.But immune regulation function of AMSCs on psoriasis vulgaris patients for PBMC was weaken.Two cell groups slightly suppressed Th17 polarization of total lymphocytes.Future,We found that medicinal indianmulberry root polysaccharide(MOP)have no effect on the phenotypes,cellcycle,adipogenic and osteogenic capacity.While it increased the mRNA expression of TGF-??IL-10?IDO?PD-LI TLR3,and decreased the mRNA expression of TNF-??TLR4.After co-culture with PBMC.MOP slightly increased the Treg proportion of total lymphocytes and the mRNA expression TGF-??IL-10 through promoting AMSCs,but it has no significant influence the Th17 proportion of total lymphocytes and the mRNA expression IL-17?IL-23Conclusion Compared with the control,there are significant differences inpatients with psoriatic AMSCs proliferation and adipogenic differentiation ability,immune inflammation suppression ability?AMSCs on psoriasis vulgaris patients's role upon inflammatory and pro inflammatory T lymphocytes of immunomodulatory function reduced,especially inflammatory T lymphocytes.MOP have effect on pro inflammatory and inflammatory functions of AMSCs on psoriasis vulgaris patients.AMSCs on psoriasis vulgaris patients Treg proportion of total lymphocytes were increased.And the levels of inflammatory cytokines in PBMC were increased,and the levels of pro inflammatory cytokines in PBMC were decreased...
