Objective:Psoriasis is a chronic inflammatory skin disease which is characterized by erythema and scale, and its etiology and pathogenesis is completely complex. Present studys show that the disease is a T-cell dysfunction immune disease controlled by polygenic inheritance background under the influence of environmental factors and T-cell dysfunction is the central links to maintain disease activity, which is closely related to psoriatic lesion occurrence, development and continuance. Involved in the pathogenesis of psoriasis, Thl/Th2cell imbalance acting on psoriasis occurrence is still being paid close attention to in addition to recent hotspot T helper cell17(Thl7cells) and so on. Among them, Interleukin-25(IL-25) and Thymic Stromal Lymphopoietin (TSLP) have been playing important roles in regulating the balance of relevant T-cells. The following are preliminary discussed in this study:1.The expression levels of IL-25and TSLP mRNA and difference between patients with psoriasis vulgaris and normal control group in peripheral blood;2The expression difference of IL-25and TSLP mRNA between the patients of different genders;3.Correlation between the expression levels of IL-25and TSLP mRNA in peripheral blood and Psoriasis Area and Severity Index (PASI);.4. Expression level correlation between IL-25and TSLP mRNA to explore the role of the two cytokines in the pathogenesis of psoriasis vulgaris;5The changes of expression levels of IL-25and TSLP mRNA in peripheral blood after four-week and eight-week treatment by total glucosides of peony(TGP) capsules to explore therapeutic effect of TGP capsules in the patients.Methods:Real-time fluorescence quantitative reverse transcription polymerase chain reaction (RT-PCR) method was used to detect the50cases and40normal persons respectively in terms of their expression levels of IL-25and TSLP mRNA in peripheral blood. Marks were given to50cases according to the PASI score standard. Using statistical software to analyze the correlation between the expression levels of IL-25and TSLP mRNA and the PASI score and P<0.05was considered statistically significant.Results:1.The expression levels of IL-25and TSLP mRNA in peripheral blood of patients with psoriasis vulgaris was obviously lower than those of the normal control group (5.21±1.31vs2.50±1.39, t=-9.47, P<0.001;3.65±0.77vs3.32±0.93, t=-2.20, P=0.031).2. The expression levels of IL-25and TSLP mRNA in peripheral blood of the patients of different genders showed no otherness (both P>0.05).3.The expression levels of IL-25and TSLP mRNA in peripheral blood of the patients both showed negative correlations with PASI score (*r=-0.366,*P=0.009;**r=-0.581**P=0.010).4. The expression levels between IL-25and TSLP mRNA in peripheral blood of the patients were positively correlated and statistically significant(r=0.452, P=0.001).5. After four-week treatment by TGP capsules, the PASI score decreased and the expression levels of IL-25and TSLP mRNA in peripheral blood of the patients were higher than they were before the treatment, and all of them were statistically significant (5.24±1.42vs3.13±0.82, t=8.37,P<0.001;3.90±0.56vs3.44±0.63,t=1.84, P=0.043;9.4±4.2vs5.8±2.7, t=4.68,P<0.001); After eight-week treatment, the expression levels of IL-25and TSLP mRNA in peripheral blood of the patients were higher than they were after four-week treatment while the PASI score decreased, and all of them were statistically significant (3.44±0.41vs2.69±0.56, t=4.71,P<0.001;3.61±0.51vs3.34±0.70,t=2.38, P=0.026;7.3±2.5vs3.8±1.8, t=5.47,P<0.001).Conclusion:1. The expression levels of IL-25and TSLP mRNA in peripheral blood of patients with psoriasis vulgaris were lower than those of normal control group while they both elevated after the application of TGP and showed negative correlations with PASI score, which indicated that IL-25and TSLP might have an inhibitory effect on psoriasis vulgaris to some degree.2. The PASI score obviously decreased and the expression levels of IL-25and TSLP mRNA elevated in the patients after four-week and eight-week treatment by TGP, which proved that the efficiency of TGP on psoriasis vulgaris was exact, while TGP might not play a role in adjusting the balance of related cytokines through IL-25and TSLP.
