Objective In this study,we treat mice model of irritable bowel syndrome(IBS)with resveratrol to observe whether the animals performance of anxiety and depression will be improved;By detecting the intestinal motility and visceral sensitivity,we try to evaluate whether resveratrol may become a potential safe and effective drug of for therapy for IBS.Method 1.Chronic and acute combining stress mice model(CACS)with IBS were established with chronic unpredictable mild stress and acute wrap restrain stress.Ninety-six rats were divided randomly into eight groups(n=12):normal group?CACS model group?resveratrol-treated(RES)group(2.5?5?10 mg/kg of resveratrol via intragastric administration)and three positive drug control groups.All rats were given behavioral tests,including sucrose consumption test,open field test,the marble burying test and elevated plus maze test to detect their anxiety and depression-like behavior.The anxiety of rats was observed and measured in sucrose consumption test,open field test,the marble burying test and elevated plus maze test,and then the visceromotor sensitivity were assessed by abdominal withdrawal reflex(AWR),intestinal motility were valued by Pigment propulsion test.Results CACS IBS rat model Showed a significant depression as the extension of time,and all of the three different dose RES groups can improve the depressive symptom.IBS model mice all showed high anxiety,resveratrol(10 mg/kg)relieved the depression performance.In the marble burying test,IBS model rats showed anxiety,and RES group(2.5?5?10 mg/kg)can reduce the performance.Intestinal viscera sensitivity was obviously elevated in the IBS model group after given large quantity of air through balloon dilating rectum and the three groups resveratrol(2.5?5?10 mg/kg)can reduce the intestinal viscera hypersensitivity.In the aspect of Intestinal motility,IBS model rats showed weakened intestinal motility,resveratrol(2.5?5 mg/kg)can reverse this status remarkably.These results suggested that IBS model mice present mood alterations such as depression and anxiety,also accompany with intestinal visceral hypersensitivity and decreased intestinal motility,resveratrol can reverse those changes.This research provides the experimental basis to the use of resveratrol as a safe and effective dual-target effect of brain and intestine for the treatment of IBS.Conclusions CACS IBS mice model present mood alterations such as depression and anxiety,also accompany with intestinal viscera hypersensitivity and intestinal motility change.Resveratrol can reverse the symptoms of anxiety and depression in irritable bowel syndrome mice and improve intestinal hyper-sensitivity and intestinal motility.
