[Background and Objectives]Non-small cell lung cancers (NSCLC) accounts for about 80% of lung cancers which are the most common type of malignancy in China, and we have seen an upward trend in incidence over the last few years. Chemotherapy serves as the main treatment for mid-stage and advanced NSCLC, to which the objective response is only around 21% or even lower. It has been found that the ?-tubulin? gene located at 6p21.3 has four exons, and the fourth exon is the most susceptible to mutations in the binding site. The gene mutation can change the stability of the microtubules,leading to weaker interaction between drugs such as paclitaxel and Vinorelbine and binding sites, or even causing the drugs ineffective. Therefore, to exploreXuanwei sensitive crowd, selecting appropriate treatment object, reducing the occurrence of taxol drug resistance is a big problem urgently to be solved in lung cancer chemotherapy.Results published today by research institutions at home and abroad exhibit great difference in showing the association between the expression of ?-tubulin ? in tumor tissues and the sensitivity to paclitaxel or vinorelbine, and it is unclear whether such difference is related to the regional differences in the target population. Although the appropriate genetic studies on the effectiveness of some chemotherapy drugs have been carried out in a few places in Beijing, Shanghai, Guangzhou and other regions,most of these research institutions have only focused on detection of local lung cancer patients and the results vary greatly with the reported clinical efficacy of the drugs used to treat lung cancers. This project was to detect the expression of ?-tubulin ?in Xuanwei lung adenocarcinoma and the sensitivity to paclitaxel, explore the effective treatment strategy to deal with high incidence of lung cancer in Yunnan Province and propose a specific comprehensive treatment plan for lung cancer in the region, which would be of great social and scientific significance in improving the clinical efficacy of lung cancer in this region.[Methods]1. It is differential to ?-tubulin ? expressing in Xuanwei lung adenocarcinoma and lung adenocarcinoma A549(1) To detect of ?-tubulin ? expression in Xuanwei lung adenocarcinoma and lung adenocarcinoma A549 by immune histochemical method.(2) It is selecting the five different areas under optical microscope in 200 times that to calculate positive cell of ?-tubulin ? expressing in Xuanwei lung adenocarcinoma and lung adenocarcinoma A549 .(3)To detect expressing of ?-tubulin?protein between Xuanwei lung adenocarcinoma and lung adenocarcinoma A549 by western blot.2. It to effect of paclitaxel in Xuanwei lung adenocarcinoma and lung adenocarcinoma A549 of ?-tubulin? differential expressing.To detecting paclitaxel concentrations with time migration influence cell biological activity in Xuanwei lung adenocarcinoma and lung adenocarcinoma A549 by MTT method.3. The statistical methodsUsing SPSS 17.0 statistical software for data analysis, comparing the mean differences between the two groups using single factor analysis of variance and independent samples t test, P< 0.05 for the difference was statistically significant.[Results]1.In the immunohistochemically experiments: average expression rate of ?-tubulin?is 95%in Xuanwei lung adenocarcinoma cancer cells, average expression rate of?-tubulin?is86%in lung adenocarcinoma A549 cell.2. In Western blot experiments: through the analysis of the Image J ,the average grey value is 1.196 in Xuanwei lung adenocarcinoma cancer cells of ?-tubulin?expression, the average grey value is 0.912in lung adenocarcinoma A549 cell 1 of?-tubulin?expression.3. In MTTexperiments: Theinhibition rate of lung adenocarcinoma A549 cells increased with the increase of paclitaxel concentrations increased, when the taxol concentration 25 ug/mL its average inhibition rate was 93%,the concentrations of paclitaxel 50 ug/mL, on average, the inhibition rate was 95%.Conditions, at the same concentration within 24 h, 48 h, 72 h compared with the passage of time between the inhibition rate increased (as shown in figure 2-1). The inhibition rate of Xuanwei lung adenocarcinoma cancer cells increased with the increase of paclitaxel concentrations increased, when the paclitaxel concentration was 25 ug/mL ,the average inhibition rate was 52%, the concentrations of paclitaxel 50 ug/mL, the average inhibition rate was 90%. At the same concentration under, the inhibition rate increased when they compared each other between 24 h, 48 h and 72 h go along with time(as shown in figure 2-2).At the same time with concentration under, average inhibition rate of paclitaxel is higher on lung adenocarcinoma A549 cells than Xuanwei lung adenocarcinoma cancer cells (as shown in figure 2-3 to figure 2-5).[conclusion]1. ?-tubulin? protein expressing higher in Xuanwei lung adenocarcinoma and there may be a regional.2. It showed the resistance to paclitaxel in ?-tubulin? high expression of Xuanwei lung adenocarcinoma.3. The ?-tubulin? protein expression level is closely related to the sensitivity of paclitaxel in Xuanwei lung adenocarcinoma, it can be used as a predictor based on paclitaxel chemotherapy drug curative effect.
