[Objective]:Studying the expression level of beta microtubules protein-IIIfrom non-small cell lung cancer,discussing the relations between the clinical and pathological features of patients and the expression level of beta microtubules protein-III from non-small cell lung cancer,discussing the relations between the clinical effect with the patient and the expression level of beta microtubules protein-III from non-small cell lung cancer[Method]:1. During March1,2007-March30,2009,we can apply the digital medical record retrieval system of the third affiliated hospital of Kunming medical Univercity to collect83patients with NSCLC. from chest surgery and cardiovascular surgery who postoperative line4cycle more the west he cisplatin chemotherapy scheme. Establishing patients detailed records, the main content including gender, age, the clinical and pathologic stage classification, smoking history, histology and lymph node is transfer, etc.2. In the pathologists department of the third affiliated hospital of Kunming medical Univercity we can retrieve83patients with NSCLC cancerous tissue and cancer tissue adjacent to wax piece who meets the conditions, and the same period which do the surgical treatment of20cases of lung benign lesions in the organization of wax. Immunohistochemistry methods were used to test beta tubulin-III protein expression level. In yunnan province cancer institute specimens library which meet the conditions of the83patients with NSCLC cancerous tissue and cancer tissue adjacent to wax piece, and the same period which do the surgical treatment of20cases of lung benign lesions in the organization of wax, the application of the real-time quantitative polymerase chain reaction method to detect beta tubulin-Ⅲ mRNA expression level.3. Analyzing the relationships between gene or protein expression level of the beta tubulin-Ⅲ and the clinical and pathological features (gender, age, the clinical and pathologic stage classification, smoking history, histology and whether lymph node metastasis), and analyzing the survival of the relationship.[Results]:1. Through immunohistochemistry methods, the examination results show the expression of B-tubulin-IIIin the cytoplasm of tumor cells, appearing clear yellow particles for positive cells colouring. Detection NSCLC cancerous tissue83cases,56cases of beta tubulin-Ⅲ express positive, positive expression rate is67.45%; Detection by83patients with NSCLC cancer organization,34of them an example beta tubulin-Ⅲ express positive, positive expression rate is40.96%; Detection lung benign lesions organization20cases,7cases of beta tubulin-Ⅲ express positive, positive expression rate of35.00%. The B-tubulin-Ⅲ expression of NSCLC tissues is higher than the B-tubulin-III expression of normal lung tissue and paracancerous tissues (P<0.05) Using RT-PCR method, The average of△Ct of the lung cancer group is2.66±0.77, beside the average train of cancer△Ct is3.99±0.79,benign lesions group for4.52±1.06.Lung cancer group compared to the average of the cancer group2-△△C for5.44±1.12,lung cancer group compared to the average benign lesions group2-△△C for2.17±0.59, the cancer group compared to the average benign lesions group2-△△C for2.88±0.93theB-tubulin-Ⅲ expression of NSCLC tissues is higher than the β-tubulin-Ⅲ expression of normal lung tissue and paracancerous tissues (P<0.05) .2. The IMH and RT-PCR method to detect,It has statistically significant between the protein or gene expression of beta tubulin-Ⅲ and pathologic stage and TNM staging relevant. It has no statistically significant between the protein or gene expression of beta tubulin-Ⅲand age, sex, smoking history, histology, lymph node metastasis.3. The rate from the patients with the exprssing positive of B-tubulin-Ⅲwas14.28%after3years of survival.Patients with negatie was37.04%after3years of survival rate. The results suggest that survival curve distribution difference was statistically significant both groups of patients, Log-rank method test statistics X2=3.912, P=0.048<0.05.[Conclusion]:1. The positive expression rate of beta tubulin-Ⅲ gene expression of protein in NSCLC cancerous tissue is higher than lung cancer tissue adjacent and the benign organization, which could indicate the process of the cancer in a certain extent.2. The protein or gene expression level of Beta tubulin-Ⅲ would heighten with the organization differentiation degree lower,it would heighten with the TNM staging heighten,which could play a certain role in the occurrence and development of the cancer.3. It has no statistically significant between the protein or gene expression of beta tubulin-Ⅲand age, sex, smoking history, histology, lymph node metastasis.4. The patients with the negative expression of beta tubulin-Ⅲ were better than the ones with the positive expression of it for the prognosis,which could be a important factor that could judge the prognosis of lung cancer.
