Effects Of Qigui Granule On BCL-2, BAX And PKC In Rats With Hypothyroidism

Objective: This experiment mainly through observing the influence of self-made prescription Qigui granule on BCL2,BAX and PKC in rats with hypothyroidism,in order to explain the therapeutic effects on thyroid function in hypothyroid rats,and to explore its mechanism.Methods: This experiment selected 60 healthy Wistar rats and randomly divided into 6 groups: blank group,model group,western medicine group,low dose group,middle dose group,high dose of Chinese medicine group.Except the blank group were given normal saline,other groups were given 0.1%PTU normal saline by gavage for 2 weeks to establish animal model.After the success of the model,the treatment group were given with western medicine and Qigui granules by gavage for 8weeks,then to detect the FT3,FT4,TSH and BCL-2,BAX,PKC of each group.Results: After treatment,the symptoms of hypothyroidism rats were improved in different degrees.The FT3 and FT4 values in the western medicine group and the traditional Chinese medicine group were higher than that in the model group,and there was a significant difference(P<0.01);The expression of BCL-2 in the high dose group,low dose group and Western medicine group was higher than that in the model group,and there was a significant difference(P<0.01);The high dose group,middle dose group and Western medicine group of BAX were lower than the model group,the difference was significant(P<0.05);The PKC value of Chinese and Western medicine group was lower than that of model group,and the difference was significant(P<0.01).Conclusion: 1.Qigui granule can rise apoptosis factor BCL-2 and reduce the BAX in hypothyroid rats;2.Qigui granule can reduce activation of protein kinase C in hypothyroid rats.3.Qigui granule can improve the general state of the rats with hypothyroidism,and rise FT3 and FT4,reduce the level of TSH.
