Grain-Shaped Decocting Pieces To Hypothyroidism Rats Experimental Study Of IL-6 And TNF-?

Objective:By animal study,we will observe the effects of QiGui grain-shaped decocting pieces on the level of FT3,FT4,TSH,IL-6,TNF-? of hypothyroidism rats and discuss its regulation of hypothalamic-pituitary-thyroid axis and cytokines.Methods:Sixty Wistar rats are included in this experiment.Ten of them are picked out by random:the blank group.In addition to the rats of the blank group,the other rats are made into hypothyroidism model by propylthiouracil for 15 days.After 15 days,that the level of FT3,FT4 is lower than the blank group and the level of TSH is more than 10uIU/ml means the rats have hypothyroidism.Then the fifty hypothyroidism rats are divided into 5 groups by random:the model group,the western medicine group,the Chinese medicine group(low dose,medium dose,high dose of QiGui grain-shaped decocting pieces),then the rats will receive corresponding drugs for 8 weeks.During the experiment,we will measure the weight of rats to adjust the dose of therapy medicine,observe the general appearance of the rats every week and measure the food-intake and the water-intake every 2 weeeks. After 8 weeks therapy,test the serum level of FT3,FT4,TSH,A-TG,A-TPO,IL-6,TNF-? of each group.Results:1.After therapy,the general appearance of rats is improved.2.After 15 days building model,the level of FT3,FT4 of model rats is significantly lower than the blank group(P<0.01) and the level of TSH is more than 10uIU/ml.After 8 weeks therapy,the level of FT3,FT4 of therapy groups is higher than the model group(P<0.05),the level of TSH of therapy groups is significantly lower than the model group(P<0.01).3.There is no difference of the level of A-TG and A-TPO among six groups(P>0.05).4.After therapy,the level of IL-6 and TNF-? of the model group is significantly higher than the blank group(P<0.01).The level of IL-6 and TNF-? of the western medicine group and the Chinese medicine low dose group has no difference with the model group(P>0.05),but the level of IL-6 and TNF-? of the Chinese medicine medium dose,high dose group is lower than the model group(P<0.05).Conclusions:Qi Gui grain-shaped decocting pieces can improve the situation of hypothyroidism rats,regulate the fuction of hypothalamic-pituitary-thyroid axis,upgrade the level of FT3,FT4,and degrade the level of TSH.It also can reduce the level of L-6 and TNF-?,but the dose must be higher.
