| Eucommia Eucommia ulmoides Oliv. Was gutta secondary species, also known as Si Xian, Xian Zhong, cotton, silk and jade tree Sipi, is expensive medicines our specialty. Eucommia bark and leaves are contained in a "Chinese Pharmacopoeia." Country in the early Eastern Han Dynasty have documented the use of Eucommia. In view of the medicinal and health value Eucommia, China also accelerated the pace of the comprehensive development of gutta-related products, and gradually to industrial development. This paper reviews the chemical composition and pharmacological activity of gutta, comparing different process parameters on the gutta series of wine chlorogenic acid, geniposide, geniposidic acid content Aucubin optimize the preparation of gutta series of wine processes. Established by HPLC method Eucommia Leaves chlorogenic acid, geniposide, geniposidic acid content compared levels of three different varieties of Eucommia leaf active ingredients. But also on the content of flavonoids in the leaves of six different species were determined gutta intended effects of different varieties of health value of Eucommia leaf; Eucommia leaf separation and purification of chlorogenic acid were studied. This will provide a theoretical basis for the development and application of gutta-related products. In this paper, results are as follows:Firstly, the establishment of a series of wine by HPLC Eucommia chlorogenic acid, geniposide, assay methods Geniposidic acid is optimum chromatographic conditions:Column:Thermo Hypersil Gold C18(250mm×4.6mm×5μm); methanol-water-acetic acid (15:85:1) solution as the mobile phase gradient elution, etc.; column temperature:25℃, flow rate:1.0mL/min; detection wavelength:237nm. The results showed that there are differences in the content of secondary metabolites in three different series of Eucommia wine. May be associated with different ratios and process parameters.Secondly, the determination of gutta series Aucubin wine by high performance liquid chromatography to study the impact of different methods of gutta liquor ratio of Aucubin content. The results showed that:with different proportions and alcohol treatment methods for content gutta Aucubin influential.Thirdly, establish a RP-HPLC method for simultaneous determination of Eucommia leaves six flavonoids (quercetin-3-0-glucose (1→2) glucoside and quercetin-3-0-arabinose (1→2) glucoside, quercetin-3-0-glucoside, isorhamnetin-3-0-β-D-glucoside, AST, quercetin) content approach. Determine the optimum chromatographic conditions are:the use of Thermo HYPERSIL C18(BDS250mm×4.6mm×5μm) column, gradient elution, detection wavelength360nm, column temperature was25"C, the flow rate of1.0ml/min. Under this condition, respectively Eucommia leaves of different varieties to determine the content found Eucommia leaf flavonoid content in these six different varieties vary greatly. Eucommia leaves maximum of six flavonoids,1.46%. Also on Eucommia leaf secondary metabolites in three different varieties were determined. Provide a theoretical basis for the development of gutta series of wine.Fourthly, Chlorogenic acid purification process:the use of50%ethanol was heated to reflux extraction, solid-liquid ratio1:10, PH=2, reflux2times per30min. The extract was through active carbon, after a1%solution of chitosan flocculation using HPD100macroporous resin adsorption, with50%ethanol elution was carried out at a flow rate2.0mL/min, each collecting100mL, purified liquid and then through active carbon After freeze-dried to give22%of the crude product, and then ethyl acetate was36.8%chlorogenic acid crude. |
