Purpose:Creating a SD rat mercury animal model by Gavaging mercuric chloride.To study the injury of SD rats' vascular endothelial cell poisoned with different doses of HgCl2.Providing the evidence that mercury directly hurt vascular endothelial cel,And analysising the level of hurt,Mercury poised is relevant to Cardiovascular Diseases.Methods:1.The SD Rat animal model is created by gavaging different does HgCl2,one of the successful judging standard is the symptom of nerve and renal injury,the other one is the level of mercury value in Blood.2.Animal grouping:A total of 54 male rats were randomized into 4 groups,high,medium,low doses of HgCl2 poisoned group with 15 in each,and 9 in control group.3.Monnitoring index:The levels of Hg in blood,NO and ET-1 in serum were measured on the 7th day,14 th day and 21 th day,the number of circulating endothelial cells(CEC)and vascular intimal thickness of upper abdominal aorta was counted at the same time.Backgroud: Mercury and its derivative: methyl-mercury have a huge damage for human health and ecological environment,due to its migration,persisitence,high bioconcentration and high biological toxicity.It is always regarded as a big subject for Environmental Medicine study.Mercury may be an important independent risk factor for cardiovascular and cerebrovascular diseases,in 2000's Academic report.In abroad,it is proveded out that the function of Vascular endothelial cells will be impacted in the Mercury environment,now we hypothesis that Mercury make a damage to vascular endothelial cell,but there are lack of reliable research in mechanism of injury.Meanwhile the related studies are blank in our country.We have a plan for this study,after created a SD rat mercury animal model,we will observe body's related changes,and study the possible mechanism of injury of Vascular endothelial cells.Results:Level of Hg: The levels of Hg in control group are close to zero,there are statistical differences between toxic group and control group(14thd:Phigh=0.010;21thd:Pmedium=0.029;Plow=0.016).Levels of serum ET-1: Comparison between groups,there are statistical differences between high dose group and control group(21thd:Phigh=0.010).Comparison in group inside,there are statistical differences during different time(high dose HgCl2 group 7thd VS 14 thd,P=0.010).Levels of serum NO: Comparison between groups,there are statistical differences between toxic groups and control group(21thd:Phigh =0.010,Pmedium =0.016,Plow =0.016).Comparison At the same time,there are statistical differences in different dose groups(7thd: high dose HgCl2 group VS low dose HgCl2 group P=0.016).Comparison in group inside,there are statistical differences during different time(high dose HgCl2 group,7th day VS 14 th day P=0.009;medium dose HgCl2 group,14 th day VS 21 th day P=0.008;7th day VS 21 th day P =0.032;low dose HgCl2 group,7th day VS 14 th day P=0.016;7th day VS 21 th day P=0.016).Circulating endothelial cell counts: Comparison between groups,there are statistical differences between toxic groups and control group(14th day:Phigh=0.024,Pmedium=0.036,Plow=0.393;21th d:Pmedium=0.036,Plow=0.036).Comparison in group inside,there are statistical differences during different time(high dose HgCl2 group: 7thd VS 14 thd P = 0.004;medium dose HgCl2 group: 7th d VS 14 thd P = 0.036,14 thday VS 21 th day P = 0.008,7thd VS 21 thd P = 0.036;low dose HgCl2 group: 7thd VS 14 thd P = 0.413,14 thd VS 21 thd P=0.008,7thd VS 21 th d P = 0.016).Conclusions:It is proved that SD rat mercury animal model is created Successfully by Gavaging mercuric chloride.HgCl2 can cause direct injury and dysfunction to vascular endothelial cells and its injury has the same changing trend with dose and time.But the injury can not directly make the vascular endothelium thickening.
