Effects of Electroacupuncture at Tinggong and Yifeng onmiR-188 and Nrp-2 Expression in Dorsal Cochlear Nucleus ofGuinea Pig Following Chronic Kanamycin-induced Deafness Objective: To explore effects of electroacupuncture at Tinggong and Yifeng on expressions of mi R-188 gene and its target protein Nrp-2 in dorsal cochlear nucleus(DCN)of guinea pig following chronic kanamycin-induced deafness.Methods: 1.65 adult guinea pigs were randomly divided into three groups,including control group(CG,n=5),kanamycin treatment group(KTG,n=30)and kanamycin sulfate and electroacupuncture at auricular point group(KEG,n=30).The KTG and KEG were divided into 6 groups,including 1,7,14,28,56 and 140 days group.2.Guinea pigs were treated with kanamycin by subcutaneous injection at 500mg/kg·day in KTG and KEG,and injected with saline at the same dose of kanamycin for 7 days in CG.Moreover,guinea pigs were electroacupunctured at Tinggong and Yifeng for 15 minutes half an hour after kanamycin treatment in KEG.3.Expressions of miR-188 and Nrp-2 of DCN were detected respectively by real time quantative PCR and Western blot.4.Statiscal methods: Data was analyzed by One-Way ANOVA test and t-test was used to analyze the difference between two groups using SPSS17.0software.The results were showed as ` x±s and P<0.05 means statiscal significant.Results: 1.There were significant effects of both kanamycin and electroacupuncture on expressions of mi R-188 and Nrp-2 in DCN.2.There was a significant decrease of miR-188 expression and increase of Nrp-2 expression in the KTG at 1 and 56 days(P<0.05,compared with the CG).There was a significant increase of miR-188 expression at 1,14,56 and 140 days and decrease of Nrp-2 expression in the KEG at 1,7,14,56 and 140 days(P<0.05,compared with the KTG).Conclusions: 1.MiR-188/Nrp-2 signaling pathway may play a role in the process of kanamycin-induced deafness.2.Electroacupuncture at Tinggong and Yifeng may regulate the plasticity and promote the rebuilt of DCN through mi R-188/Nrp-2 signaling pathway following chronic deafness.Part ??
