Object: The main varities of Fructus amomi,in the present market were the Amomum villosum Lour.planted in Yunnan province(YCS)and Fructus Amomi planted in Myanmar(JKS),respectively.In this paper,the chemical compositions and the main pharmacodynamics experiments including whole animal,isolated gastrointestinal muscle caused the impact to Gastrointestinal motility,digestive juice secretion,and other experiments of analgesia were designed and employed to evaluate the differences of therapeutic material basis and the pharmacologic activities of the two kinds of Fructus Amomi,This study can provide the experimental basis for the clinical application and in-depth development of YCS.Methods: 1.The chemical constituents of YCS and JKS were analyzed by GC-MS,LC-Q-TOF-MS / MS,HPLC,phenolic phenol method and ultraviolet-visible spectrophotometry.2.The effects of two kinds of Fructus Amomi on gastrointestinal hormones such as gastric juice and Motilin(MTL),Substance P(SP)and Vasoactive intestinal peptide(VIP)were measured by using pyloric ligation.And the effects of two kinds of Fructus Amomi on gastrointestinal motility were analyzed by the experiments of dextran blue color and isolated gastrointestinal smooth muscle.The anti-diarrhea and the analgesia effects of two kinds of Fructus Amomi on mice were tested with the methods of and diarrhea induced by senna and writhing test.Results: 1.The chemical compositions of YCS and JKS are significantly different.Volatile oil content of YCS was about 3.75%,and 41 kinds of Volatile chemical compositions were detected from YCS.The highest content of volatile oil of YCS is bornyl acetate(73.61%),which accounted for 2.76% of its dry plant.And followed by camphor(17.27%);Volatile oil content of JKS was about 1.97%,and 50 kinds of Volatile chemical composition were detected from JKS.JKS containing volatile oil about 56.77% camphor,followed by acetic acid methyl ester(23.17%),and acetic acid borneol ester accounted for 0.46% of its dry plant.In addition to acetic acid borneol,camphor,borneol,D-limonene,isopuleol,?-terpinene,camphene,?-side bene,?-myrcene,?-phellandrene,the total content of vanillic acid,total phenolic and total flavonoids Quercetin-8-O-?-L-[rhamnopyranosyl]-3-O-?-L-[rhamnopyranosyl-(1'-2'')-?-D-glucopyranoside](use M754 said)in YCS are higher than that of JKS.The content of quercetin and Methyl-6-(6-(3,4-dimethoxy)-benzalacryl-?-D-glucosyl)-O-?-D-glucopyranoside(use M546 said)and protocatechuic acid in JKS was higher than that in YCS.2.The high dose of YCS and JKS could significantly inhibit the gastric secretion of the pyloric ligation rats,decrease the acidity of gastric acid and the expression of VIP and the effect of nonvolatile components,increase the activity of pepsin and the content of MTL and SP.Compared with the effect of the JKS' extract,the effect of YCS' is better.In addition,JKS non-volatile components could not reduce the role of VIP content,and its low-dose group can inhibit pepsin activity.3.In the study of gastrointestinal motility,thechemical components of volatile oil of the two plant can significantly increase the gastric residual rate.And non-volatile components of YCS also have significantly increase the gastric residual rate,but non-volatile components of JKS components have no effect on the gastric residual rate.The volatile oil of the two plants s could significantly promote the intestinal advancement.Compared with the IKS,the YCS' effect is slightly better.However,The effect of the non-volatile components of the two plants on the intestinal propulsion is not significant.The results of isolated intestinal and gastric smooth muscle of SD rats showed :(1)With the dose gradually increasing,YCS inhibited the normal intestinal muscle,JKS was inhibited-excitement-inhibition;for gastric smooth muscle,YCS and JKS non-volatile components were excited after the excitement,JKS volatile oil was inhibited-excitement-inhibition-effect(2)the two drugs on atropine caused by intestinal and gastric smooth muscle relaxation has a significant antagonistic effect,and YCS' role is slightly better than JKS.(3)the two non-volatile components of acetylcholine-induced intestinal muscle spasm has a significant inhibitory effect,volatile oil has significantly inhibited acetylcholine-induced gastric smooth muscle spasm,YCS' effect is slightly better than JKS;(4)two volatile oil on barium chloride The YCS non-volatile components,JKS volatile oil on barium chloride-induced gastric smooth muscle spasm has a significant relaxation effect,JKS is slightly better than YCS.4.The two extracts can reduce the diarrhea induced by senna,the role of YCS was significantly better than JKS,and can extend the row of thin urine latency.The two drugs can significantly reduce the number of wrinkles in mice caused by acetic acid pain,and show no difference;the volatile oil of YCS and JKS have a certain extension of the role of writhing latency,but non-volatile components of JKS has no effect.Conclusion: 1.The chemical compositions of the two plants were quite different.The content of volatile oil and the content of methylprednisolone in YCS was much higher than that of JKS,and the content of methyl acetate in JKS was less than that required by pharmacopoeia.2.YCS and JKS can reduce the amount of gastric juice and reduce gastric acidity,improve pepsin activity,and improve pyloric ligation of gastric mucosal injury in rats;can increase the excitatory gastrointestinal hormone MTL,SP concentration,reduce the inhibitory gastrointestinal hormone VIP Concentration,and showing good gastrointestinal motility,and the other can inhibit gastric emptying,promote intestinal propulsion,the role of isolated intestinal,gastric smooth muscle and drug concentration and smooth muscle function status,so the two drugs on gastrointestinal motility A significant two-way regulatory role;the other two drugs on the gastrointestinal tract showed significant diarrhea,analgesic effect.The effect of volatile oil in the above experiments is generally stronger than that of nonvolatile components.JKS showed similar effect to YCS in gastrointestinal function regulation and disease treatment,but YCS pharmacological effect is superior to JKS.Based on the significant differences in chemical composition and content of YCS and JKS,and the obvious advantages of YCS in the pharmacological action,it is suggested that the differences between the two kinds of Fructus Amomi should be given full consideration in order to obtain safe and effective Fructus Amomi Herbs.At the same time,we should increase the cultivation and application of YCS.Thus,the qualified products of Fructus Amomi could be obtained in pharmacological and food fields.
