Objective:Chunxuan No.1-F4 was obtained by traditional farm cultivars of Amomum viliosum Lour, called Longfruit No.2 (female parent) and Amomum longiligularg T. L. Wu (male parent) and after 4 generations of breeding. The prophase work has studied on the germplasm characteristics, plant yield and molecular conformation, but its effective ingredients and contents of volatile oil were lack, moreover it was lack of pharmacological experiment evidence. Thus, in this study the content of volatile oil, bornyl acetate content and volatile oil components and relative contents of Chunxuan No.1-F4 would be determined, and the pharmacological of Chunxuan No.1-F4 volatile oil was researched according to the clinical indications of Amomum villosum. In addition, immunoregulatory activity of polysaccharide in Chunxuan No.1-F4 was studied. To enrich and improve the quality evaluation of Chunxuan No.1-F4, in order to solve the problem of Amomum villosum natural pollination, lay the foundation of expanding Amomum villosum germplasm resources.Method:1. The differences of volatile oil and effective composition in Chunxuan No.1-F4 and its female parent Longfruit No.2 were analyzed by steam distillation method, GC and GC-MS method. The differences on the content of effective components of the volatile oil components were studied. Explore the properties of new hybrid seed germplasm from a chemical perspective.2. Used Senna induced mice diarrhea model, Montmorillonite Powder as positive drug, administered 10% Senna decoction, observed and record the mice wet faeces number in 4 hours, t of volatile oil in ChunxuanNo.1-F4 and Longfruit No.2 on mice diarrhea. To investigate the impact of Chunxuan No.1-F4 and Longfruit No.2 volatile oil on mice diarrhea.3. Established mice auricle swelling model induced by xylene, Dexamethasone Acetate tablets as positive drug, calculated the ear swelling degree and average and ear swelling inhibition rate; investigated the effect of Chunxuan No.1-F4 and Longfruit No.2 volatile oil on mice auricle swelling.4. Established mice pain model induced by acetic acid solution, the Rotundine tablets as positive drug, to investigate the effects of volatile oil from Chunxuan No.1-F4 and Longfruit No.2 on mice the writhing response latency and writhing pain.5. Used Spectrophotometric instrument to measuring gastric residue by absorbance value, to calculation the relative rate of residual pigment in gastric. Measure the distance from pyloric sphincter to the front of the cecum and pigment, to calculation the advance ratio of small intestine. To investigate the effects of volatile oil from Chunxuan No.1-F4 and Longfruit No.2 on mice gastric emptying and small intestine propulsion function.6. Took the isolated rabbit jejunum, and record the effects of different drugs on spontaneous activity of jejunum by biological systems, to investigate the effects of volatile oil from Chunxuan No.1-F4 and Longfruit No.2 on the isolated rabbit jejunum spontaneous activity.7. Finally, low immunity induced by cyclophosphamide in mice to investigate ACSP of immunocompromised mice immune organ weight, macrophage phagocytosis, spleen hemolysin effects, to evaluate the immunomodulatory activity of ACSP.Result:1. The volatile oil and main effective components bornyl acetate in Chunxuan No.1-F4 and Longfruit No.2 were in line with the requirements of Chinese Pharmacopoeia, but the the content of volatile oil ChunxuanNo.1-F4 was higher than Longfruit No.2, and there was significant difference between them, the content of borneol acetate in Chunxuan No.1-F4 was slightly lower than Longfruit No.2. There was slightly difference in volatile oil group between them, and both of them had 14 kinds of component.2. Both Chunxuan No.1-F4 and Longfruit No.2 volatile oil had significant antidiarrheal effect, but no significant difference between them, the antidiarrheal effect were the same with Montmorillonite powder. The volatile oil of them can significantly reduce xylene induced mouse ear swelling, the anti-inflammatory effect were equal with Dexamethasone Acetate Tablets. And the two volatile oil is equivalent inhibitory effect on xylene induced ear swelling;The volatile oil of them had significant analgesic effects, and had the same analgesic effect, and the analgesia effect can be comparable to Rotundine tablets. Both Chunxuan No.1-F4 and Longfruit No.2 volatile oil in high dose was significantly inhibited gastric emptying in mice, and low doses in promoting gastric emptying. Both of them have the bidirectional regulation effect on gastric emptying, and the effect is similar. Two kinds of Amomum villosum volatile oil in high, middle dose can promote intestinal propulsion in mice, and low dose had no obvious effect, there was no difference on the intestinal propulsive effect, and the effect is similar with cisapride. Two volatile oil can significantly inhibit the spontaneous activity of isolated rabbit jejunum, and its effect is better than that of atropine, low dose of Chunxuan Ho.1-F, volatile oil on inhibitory effect was better than Longfruit No.2.3. ACSP was able to improve clearance ability and phagocytic speed of mice peritoneal macrophage. Increase the content of serum hemolysin, thymus and spleen index, and improve the immune capacity in mice. ACSP was the same with ASP on immune regulating activity.Conelusion:The research shows that, Chunxuan No.1-F4 main chemical components were similar to its female parent, Chunxuan No.1-F4 volatile oil and polysaccharides has the same pharmacological effects with Longfruit No.2, and the activity was same. Chunxuan No.1-F4 not only has excellent germplasm characteristics, but also has the same pharmacodynamic function with Longfruit No.2, is an ideal hybrid species, it was worth to popularize. In order to solve the problem of Amomum villosum natural pollination, lay the foundation of expanding Amomum villosum germplasm resources.
