| Objective:In traditional Chinese medicine theory as the instruction,this study adopts advanced gene sequencing and real-time fluorescent quantitative PCR detection technology,through the determination of p16 gene and its mRNA expression to analyze genetic features of kidney Yin deficiency type senile patients,and using the Shouwu granule(II)to make objective evaluation about the curative effect of the kidney Yin deficiency type senescence,and further explore the Shouwu granule(?)intervention in patients with kidney Yin deficiency type senile response of individual differences and p16 gene mRNA expression,and T3?T4?TSH internal environment change,to expect further interpretations of aging mechanism with traditional Chinese medicine,and provide the basis of genetic biology.Methods:Will 60 cases of 60-80 years is in conformity with the experimental diagnostic criteria and inclusion criteria of kidney Yin deficiency aging,adopt the method of single blind,parallel-group,30 cases were randomly divided into treatment group and control group 30 cases.Two groups of subjects were taking Shouwu granule(?),Liuwei Dihuang Wan.Treatment to the patient before the random detection of p16 gene expression characteristics and spectral distribution,8 weeks after treatment compared before and after treatment the two groups of p16 gene mRNA expression change,the change of serum T3,T4,TSH,clinical curative effect,symptom integral,psychological age grading,daily life activities ability rating scale.Results:1.In the two groups before and after the treatment of traditional Chinese medicine symptom integral comparison:comparison between the two way before treatment(P>0.05,there was no statistically significant difference,that comparable;Two groups after treatment of TCM symptom integral reduced,the differences in the group P<0.01,explain differences between the two groups before and after treatment were statistically significant meaning;Score lower than the control group,treatment group after treatment compared between groups(P<0.01,with significant difference statistically significant,the treatment group is better than that of control group in improving TCM symptoms integral.2.Syndromes curative effect comparison:the two groups after treatment in treatment group and control all 30 cases,the treatment group and the control group only 2 cases were markedly effective,6 cases had marked effect,17 cases,effective cases of treatment group and control group there were 12 cases were markedly effective,ineffective treatment has 7 cases in treatment group,16 cases in the control group,treatment group total effectiveness 76.7%,control group total effective rate was 46.7%.The two groups of clinical curative effects were examined by non-parametric tests of hierarchical data,and P<0.01 showed that the two groups were statistically significant and the treatment group was superior to the control group.3.Before and after treatment the two groups of psychological ageing scores were compared:the comparison between the two groups was the comparison of P>0.05 and the non-statistical significance of the treatment.After treatment,both groups had lower ratings of aging,compared with P<0.01 in the group,which showed significant statistical significance in both groups.The comparison P>0.05 in the post-treatment group showed that there was no statistical significance in improving the psychological aging of the patient.4.The comparison between the two groups of daily living abilities was compared:the comparison between the two groups,P>0.05,no statistical difference,and the comparison;After treatment,the two groups had decreased their ability of daily living ability,compared with P<0.01 in the group,which showed significant statistical significance in both groups.Score lower than the control group,treatment group after treatment compared between groups(P<0.05,the difference is statistically significant,the treatment group in improving daily life ability is better than the control group.5.The comparison of the expression content of the mRNA in the two groups of p16 gene was compared before and after treatment:compared with P>0.05 in the pretreatment group,the comparison between the two groups was suggested.The two groups of p16 gene mRNA were decreased in the two groups,compared with P<0.01 in the group,which indicated that the variation of the mRNA expression in both groups was statistically significant.Patients in treatment group after treatment of p16 gene mRNA expression levels lower than the control group,the comparison between groups(P<0.05,the difference is statistically significant,the treatment group in reducing p16 gene mRNA expression level is better than that in control group.6.The comparison of blood T3 in the two groups before and after treatment was compared:compared with the previous group,P>0.05,which suggested that the balance of the two groups was comparable;The serum T3 levels were raised in both groups,compared with P<0.01 in the group,which showed significant statistical significance in both groups.Treatment group serum T3 was higher than the control group,compared with P<0.05 in the group,the difference was statistically significant and the treatment group was superior to the control group.7.The comparison of blood T4 in the two groups before and after treatment was compared:compared with the previous group,P>0.05,which suggested that the balance of the two groups was comparable;The serum T4 levels were raised in both groups,compared with P>0.05 in the group,which showed the difference between the two groups was not statistically significant.8.The comparison of blood TSH in the two groups before and after treatment was compared:compared with the previous group,P>0.05,which suggested that the balance of the two groups was comparable;The serum TSH levels were raised in both groups,compared with P<0.01 in the group,which showed significant statistical significance in both groups.The treatment group TSH content was higher than the control group,compared with P<0.05 in the group,the difference was statistically significant,and the treatment group was better than the control group.Conclusions:1.Shouwu granule(?)can effectively improve the clinical symptoms of TCM in patients with kidney Yin deficiency aging,and improve mental aging degree,improve the ability of daily life.2.Shouwu granule(?)can reduce the patients with kidney Yin deficiency aging p16 gene mRNA expression levels.3.Shouwu granule(?)can improve the kidney Yin deficiency aging patients serum T3 and TSH level,adjust the hypothalamus-pituitary-thyroid axis virtual excited state,restore the environment balance,then improve the clinical symptoms,reach the role of anti-aging.4.Shouwu granule(?)is safe and effective prevention and treatment of kidney Yin deficiency aging. |
