Background and objective:tooth trauma is the most common disease in oral and maxillofacial surgery.The incidence of dental trauma was gradually increased.The probability of permanent teeth trauma occurred about 16%teeth tooth injury incidence rate is about 7%-21%.And most happened in anterior teeth.Tooth trauma give patients a lot of troubles in functional and aesthetic.So,the preservation and treatment of ingury tooth is important for the preservation of the dislocated teeth.Now the best treatment to tooth truma is replantation immediately.So we can keep odontogenic cells(periodontal ligament stem cells and dental pulp stem cells)activity and make the regeneration happen again.The preservation effect of odontogenic cells and odontogenic cell activity changes with time were studied through different media we can access in daily life,in hope of finding the best kind of medium can keep cells activity to the maximum and the best medical treatment.And also guide doctors' treatment with the standardized action.Materials and methods:The healthy teeth simulating normal dislocation teeth as experimental materials,selection of healthy young people under the age of 28,impacted teeth without periodontal disease or pulpitis,ensure the active cells of periodontal ligament cells and dental pulp cells.From October 2016 to January 2017,we collected 48 impacted teeth from the age below 28,without caries and periodontal pulp of any disease.Before collected,the patients were told and agreed to take their teeth for experiment.The teeth were divided into four groups,as the positive control group,the experimental group one,the experimental group of two,the negative control group.In positive control group,the dislocated teeth were stored in the DEM medium.Experiment one:place the dislocated teeth in milk.Experiment two:place the dislocated teeth in the egg white.In negative control group,the dislocated teeth were stored in the saline.Each group was divided into 6 groups according to different time periods,as Oh,0.5h,1h,2h,4h,6h.Each group had 2 teeth.As the periodontal cells and the pulp cells was collected.Neutral enzyme digestion with 2.4mg/mL addition.After digestion,centrifugal and uniform.Then the trypan blue solution was added.After 3min,the number of living cells and dead cells were observed and counted under the light microscope.The survival rate of cells was calculated at different time periods,and the cell survival curves were plotted.Finally,make the statistical analysis.Subject characteristics:patients are all young groups,teeth were all impacted and without periodontal disease or pulpitis,periodontal cells and pulp cells were active.Totally healthy,no organ and coagulation disorders,with good compliance..Results:(1)after trypan blue staining,it was observed that the active cells were colorless and transparent spheres,the cells were plump and round,and the cytoplasm was uniform.The dead odontogenic cells uniform blue nuclei,not visible,irregular cell morphology,may also occur for circular cracking and deformation.(2)in the same time,the preservation effect of DEM medium is the best,and it can keep the cells active.The effect of milk is not as good as the DEM medium,but better than the egg white.Saline is not the proper storage medium for tooth truma.(3)the effect of periodontal cells in milk was better than the effect of periodontal cells in egg white.The preservation effect of pulp cells in milk was better than the effect of periodontal cells in egg white.(4)the activity of periodontal ligament cells in the 0.5h group was 98.60%.The activity of periodontal ligament cells in group 1h was 97.62%.The activity of periodontal ligament cells in group 2h was 96.64%.The activity of periodontal ligament cells in group 4h was 94.71%.The activity of periodontal ligament cells in group 6h was 90.44%.Before the 1h of the experiment,every half hour activity decline about 1.5%.In the time after 1h,the hourly attenuation is about 2%.The activity of periodontal ligament cells in the 0.5h group was 91.12%.The activity of periodontal ligament cells in group 1H was 82.87%.The activity of periodontal ligament cells in group 2H was 79.80%.The activity of periodontal ligament cells in group 4h was 77.49%.The activity of periodontal ligament cells in group 6h was 74.74%.In the first 1h,periodontal ligament cells decline 9%in every half hour.In the time after 1h,the hourly attenuation is about 1.5%.(5)the activity of dental pulp cells in the 0.5h group was 98.26%.The activity of dental pulp cells in group 1h was 91.63%.The activity of dental pulp cells in group 2h was 87.56%.The activity of dental pulp cells in group 4h was 78.02%.The activity of dental pulp cells in group 6h was 71.52%.Before 1h of the experiment,pulp cells death rate is about 10%,after 1h the decline of pulp cell rate is about 3%.The activity of dental pulp cells in the 0.5h group was 97.55%.The activity of dental pulp cells in group 1h was 84.77%.The activity of dental pulp cells in group 2h was 81.74%.The activity of dental pulp cells in group 4h was 77.34%.The activity of dental pulp cells in group 6h was 63.15%.The decline rate of 1h is about 13%,after the decline of the rate of dental pulp cells accelerated,the decline rate is about 3%in group 1h-4h.The death rate is about 6.6%in group 4h-6h.Conclusion:(1)milk has obvious advantages over preserved egg white in preservation of periodontal ligament cells and dental pulp cells.(2)if you can not get milk,you can also use the egg white as a storage medium,but you need fast medical treatment,because the decay rate of dental cells in the egg white is faster.(3)the rate of decline of periodontal ligament cells and dental pulp cells in milk was stable and lower than that of egg white cells.These results suggest that milk has an advantage in preserving the viability of periodontal ligament cells and dental pulp cells.
