Objective:To compare the DMP1, AMBN, Collagen-I and DLX1in the amount of difference in the relative expression of mRNA SD in the rat tooth germ development, explore SD rat tooth germ during differentiation, the relative expression of these genes volume changes, reveal the stage specific expression pattern, to explore suitable for tooth tissue engineering seed cells. Methods:Tooth germ tissue from embryonic the14day, the17day of SD of fetal and postnatal the4day in the SD rat, tooth germ cells and tooth germ tissue of the4day SD rat was born after in vitro culture was the1day, the3day, the6day were compared by real time quantitative PCR detection of relative expression changes in the amount of tooth gene mRNA levels. Results:DMP1, AMBN, Collagen-I and DLX1mRNA expression in tooth germ tissue at the4day after birth and embryonic the17day were higher than that of embryonic the14day, the difference was statistically significant (P<0.05). Relative expression of tooth germ cells in DMPl,AMBN,Collagen-I genes mRNA in cultured the3day was higher than cultured for the1day and the6day, the difference was statistically significant (P<0.05), and the relative expression of DLX1mRNA in cultured for the1day was higher than those cultured for the3day and the6day, the difference was statistically significant (P<0.05). Conclusion:The tooth germ tissue the4day after birth in four teeth of relative genes expression data can be used as seed cells for more. In the3day after cultured tooth germ cell proliferation and differentiation is the good period can be used to construct tissue engineered tooth.
