Objective The aim of this study was to evaluate the influence of platelet-rich plasma (PRP) on cultured periodontal ligament fibroblasts, using an in vitro model of wound healing,and assess the effect of PRP on periodontal healing in replanted teeth in Beagle dogs. Methods 1. Platelet-rich plasma (PRP) was isolated by two-step centrifugation. Primary PDLFs were obtained from mongrel normal first premolar. The 4th passage of PDLFs were cultured in 12-well chamber plates. The cells were grown to confluence and in vitro wounds were mechanically created, removing a 8㎜ diameter cells within a circular area using a sterilized silicon bung. The wells were then incubate for 1, 4, 7, 9 and 11 days and the healing speed of these groups was evaluated. 2. The second incisors were re-implanted after 60min of dry storage . In group A and B, PRP were applied before replantation. In group A and C, the pulps were removed 7days later. All the dogs were sacrificed after 12 weeks, the histological parameters studied were heal PDL, surface, inflammatory and replacement resorption. Results 1. The proliferation and the migration of cells were enhanced by addition of 1%PRP and 10%PRP. 2. There were no significant differences between group A and C. Group B showed a lower incidence of ankylosis, and higher incidence of healed PDL compared with group D. Conclusion 1. PRP appears to exert an influence on cells that is compatible with improved wound healing. 2. Under the condition of this study, PRP could help to decrease the incidence of ankylosis and increase the incidence of healed PDL in groups that the pulps were not removed.
