ObjectiveTo investigate the Semen Litchi effective constituents(SLEC)on inflammatory cytokines transforming growth factor-β1(TGF-β1),monocyte chemotactic protein-1(MCP-1)and macrophage migration inhibitory factor(MIF)in pre-diabetes rats induced by sugar and high fat diet.Methods1.After detection of blood glucose,30 male SD rats were randomly divided into formula-1 group,formula-2 group and normal control group.The rats in formula-1 group and formula-2 group were fed with high sugar and high fat diet,and the rats in normal control group were fed with normal diet,which lasted for 20 weeks.In the 0th,10th and 20th week,orbital venous blood samples were obtained after fasting for 12 hours.The detection of fasting blood-glucose level and oral glucose tolerance test were conduct.2.60 male SD rats were randomly divided into normal control group,model group,SLEC low-dose group(3 g/kg),SLEC medium-dose group(6 g/kg),SLEC high-dose group(12 g/kg)and metformin group(0.2 g/kg).The rats in normal control group were fed with normal diet and the others were fed with the optimal high sugar and high fat diet.Drug administration lasted for six weeks and the rats in the model group and normal control group were given the same volume of saline.The contents of TGF-β1,MCP-1 and MIF in the serum of each SD rats were detected by ELISA.The mRNA expression level of TGF-β1,MCP-1 and MIF were detected by real-time fluorescent quantitative PCR.Results1.The results of the fasting blood-glucose test showed that formula-1 group and formula-2 group rats blood sugar leve were increased significantly compared with normal control group(P=0.000、0.000<0.01,P=0.000、0.000<0.01),meet the epidemiologic characteristics of pre-diabetes,there was no significant difference between the two groups(P=0.070、0.125>0.05);The results of the oral glucose tolerance test showed that rats blood sugar leve were increased significantly in the formula-1 group and formula-2 group compared with normal control group(P=0.000、0.000、0.004<0.01,P=0.000、0.001、0.000<0.01),and there was no significant difference between the two groups(P=0.766、0.067、0.697>0.05,P=0.073、0.242、0.910>0.05),it indicated that fed with formula-1 or formula-1 high sugar and high fat diet could induced pre-diabetes rat model.2.Compared with model group,fasting glucose levels were decreased significantly in the SLEC medium-dose group and high-dose group(P=0.015<0.05,P=0.000<0.01),and there was no significant difference in SLEC low-dose group(P=0.380>0.05),it indicated SLEC has hypoglycemic effects were positively related to the dose.3.Compared with model group,the levels of TGF-β1,MCP-1 and MIF in blood serum were decreased significantly in the SLEC medium-dose group,high-dose group(P<0.05),and there was no significant difference in SLEC low-dose group(P>0.05),it indicated the levels of TGF-β1,MCP-1 and MIF in blood serum were positively related to the dose of SLEC.4.Compared with model group,the expression of TGF-β1,MCP-1 and MIF in pancreatic tissues were decreased significantly in the SLEC medium-dose group,high-dose group(P=0 014、0-017、0.038<0.05,P=0.000、0.002、0.001<0.01),and there was no significant difference in SLEC low-dose group(P=0.508、0.093、0.846>0.05),the expression of TGF-β1,MCP-1 and MIF in pancreatic tissues were positively related to the dose of SLEC.ConclusionSLEC could alleviate the inflammation reaction in rats with impaired glucose tolerance,which is related to down-regulation expression of TGF-β1,MCP-1 and MIF. |
