Diabetes mellitus(DM)has many complications,diabetic gastroparesis is one of the common complications,and has a very serious impact on the quality of life of patients.At present,many studies have found that gastric emptying dysfunction caused by oxidative stress plays an important role in the pathogenesis of diabetic gastroparesis.Gastric antral smooth muscle cells are the direct source of gastric contraction and emptying.The apoptosis of gastric antral smooth muscle cells induced by oxidative stress is an important pathophysiological basis of gastric emptying dysfunction.Purpose:To investigate the role of SCF-Kit signaling pathway in the damage of ICC in gastric antrum of DGP rats and the therapeutic mechanism of Hewei Decotion.At present,modern medicine is mainly to promote gastrointestinal motility drugs to improve the symptoms of gastric emptying disorder in patients with diabetic gastroparesis,but there are not effective treatment,with a certain side effects and other issues.Hewei decoction(composed of Radix rehmanniae,ophiopogon root,polygonatum,Glehnia root,white atractylodes rhizome,bergamot,semen Raphani)is the mentor Professor Xia proposed according to the pathogenesis of DGP patients,stomach yin deficiency and spleen qi deficiency,has made a very good therapeutic effect in clinic,but the specific mechanism of drugs is not very clear.The research group found that in previous studies,which have enhanced gastric motility in diabetic gastroparesis and gastric blood flow in rats,at the same time through the upregulation of SCF-Kit signaling pathway,regulating the Cajal number of Leydig cells,improve diabetic gastroparesis rat gastric pacing function,promote gastric motility for.However,it is still necessary to verify whether the decoction can improve the level of oxidative stress and decrease the apoptosis of gastric antral smooth muscle cells.Purpose:The target of Hewei decoction in the treatment of diabetic gastroparesis rats,whether can improve oxidative stress,reduce apoptosis of gastric smooth muscle cells,thereby reversing DGP gastric emptying,provide experimental basis for the mechanism of Chinese medicine and stomach soup multi target therapy of diabetic gastroparesis.Methods:Healthy weight 201-205g adult male SD rats were randomly divided into normal control group,diabetic gastroparesis model group,model group with 70 mg/kg intraperitoneal injection of streptozotocin(1%STZ)to establish the rat model of diabetic gastroparesis.After the completion of the model,diabetic gastroparesis model group rats were randomly divided into three groups,namely the model control group,and the stomach soup group and the Mosa Billy group.Then,according to the weight were treated by Hewei decoction group and Mosapride group amount and stomach soup and Mosapride suspension gavage,the normal control group and DGP model group were given distilled water.After the administration,eyeball blood,centrifugal separation of plasma,plasma MDA and SOD content for intragastric administration of experimental gastric emptying was measured by rate of specimens from the stomach tissue,part TUNEL for detection of gastric antral smooth muscle cells apoptosis,the expression of Western blot protein extraction for detection of gastric antrum Bcl-2 protein,Bax protein and HO-1 protein.Results:1 Comparison of the generalconditionThe rats in the normal control group were soft and shiny,lively and active,sensitive and good in mental state.The rats in the model group were dull,dull,poor activity,slow reaction and poor spirit.The rats in each administration group were better than those in the model group,but were worse than those in the control group.Conclusion:Hewei Decotion can improve gastric pacing function and gastric motility by up-regulating SCF-Kit signaling pathway to regulate the quantity of ICC in DGP rats.2 The weight of each group of ratsThe weight of the rats in the normal control group continued to increase.There was no significant difference in weight between the DGP model group,the rats in the decoction group and the Mosa Billy group(P<0.05).After treatment,compared with the DGP model group,and Stomach Decoction group rats were obviously increased(P<0.05),the weight of mosapride citrate group rats increased significantly(P<0.05).Compared with the mosapride citrate group,and Stomach Decoction group rats weight loss(P<0.05).3.Comparison of blood glucose of rats in each groupAfter the administration,the blood glucose of rats in DGP model group and normal control group did not change significantly compared with the control group,the blood glucose of rats in DGP model group was significantly higher than that of normal control group(P<0.01).Hewei decoction group compared with before administration of blood glucose decreased significantly(P<0.05),but still higher than the normal level;the mosapride citrate group rats blood glucose compared with the previous administration had no significant difference(P>0.05),is still higher than the level of normal control group.4 Gastric emptying rate of rats in each groupCompared with the normal control group,the gastric emptying rate of DGP model group was significantly lower(P<0.01).After drug intervention,gastric emptying and gastric Decoction group and mosapride citrate group rats was DGP model group were increased(P<0.05);compared with mosapride citrate group rats,gastric emptying and gastric Decoction group rats was slightly lower than the mosapride citrate group,but no statistical difference learn the meaning of(P>0.05).5.Comparison of the contents of MDA and SOD in plasma of rats in each groupCompared with the normal control group,DGP model group rats and plasma MDA content of rats increased significantly(P<0.05);compared with the DGP model group,mosapride citrate group rats with Hewei decoction rats of MDA group was lower(P<0.05);compared with mosapride citrate group,and the Stomach Decoction group rats serum MDA content was significantly lower(P<0.05).Compared with the normal control group,DGP model group rats and plasma SOD content of rats was significantly lower(P<0.05);compared with the DGP model group,mosapride citrate group rats with Hewei decoction rats of SOD group were significantly higher(P<0.05);compared with mosapride citrate group,and the content of plasma SOD Stomach Decoction group were significantly higher(P<0.05).6.The changes of Bax,Bcl-2 and HO-1 protein in gastric antrum of rats in each groupCompared with the normal control group,DGP model group and Bax protein content in the medicine group were significantly increased(P<0.05);compared with the DGP model group,drug group Bax protein content decreased significantly(P<0.05);compared with mosapride citrate group,and Stomach Decoction group rats Bax protein content was relatively low,but no statistically significant difference(P>0.05).Compared with the normal control group,DGP model group and bcl-2 protein content of rats was significantly lower(P<0.05);compared with the DGP model group,drug group Bcl-2 high protein(P<0.05);compared with the citrate Mosa Billy group,and bcl-2 protein content in the Stomach Decoction group were higher,but no statistically the difference(P>0.05).Compared with the normal control group,DGP model group and HO-1 protein content in the medicine group were significantly increased(P<0.05);compared with the DGP model group,drug group HO-1 protein content increased significantly(P<0.05);compared with mosapride citrate group,and HO-1 protein content in the Stomach Decoction group were higher,but no statistical difference(P>0.05).7.Apoptosis of gastric antrum smooth muscle cells in ratsIn normal control group,gastric antrum smooth muscle cell apoptosis was less.Compared with the normal control group,DGP model group and administration group of gastric antral smooth muscle apoptosis cells increased significantly(P<0.05).Compared with the DGP model group,the apoptosis rate of gastric antral smooth muscle cells was significantly decreased in the control group(P<0.05).Compared with the mosapride citrate group,and Stomach Decoction group rat smooth muscle cell apoptosis is less,but no statistically significant difference(P>0.05)Conclusion:And by adjusting the level of oxidative stress could promote gastric body,inhibit the apoptosis of gastric smooth muscle cells,thereby improving gastric emptying in diabetic gastroparesis rats,which may be one of the mechanism of Hewei decoction in the treatment of diabetic gastroparesis.
