| BackgroundStress ulcer is an acute gastric mucosal lesion in various powerful stressors. It mainlymanifested the gastric mucosal erosions, ulcers, bleeding, and it is a clinical commondisease. Stress ulcer, especially in patients with acute bleeding, often caused the highmortality rate. Few drugs are for preventing and treatment of stress ulcer in clinical, andwe also did not know about the theoretical basis of the mechanism of these drugs clearly.Xiaotan Hewei decoction is an experience recipe in clinical practice for many years, and itis effective in treating stress gastrointestinal functional disorders. However, the relatedmechanism of Xiaotan Hewei decoction in preventing stress ulcer is still relatively scarce.This research from the perspective in stress-induced gastric mucosal cell apoptosis try toexplore the related possible mechanism of Xiaotan Hewei decoction in preventing stressulcer.It also provides us some references in the basis further research and the clinical use.ObjectiveTo elucidate the underlying mechanism of Xiaotan Hewei decoction in preventing thegastric mucosal cell apoptosis in water-immersion and restraint stress induced rats.Toestablish acute stress ulcer model in rats, and we use Xiaotan Hewei decoction to intervenethe model rats. We detect and analysis the rats’ gastric ulcer index, gastric mucosal cellapoptosis, expression of GRP78and Caspase-12in gastric mucosa tissue, eliminating theunderlying mechanism of Xiaotan Hewei decoction in preventing stress ulcer.Methods1.78male SPF healthy SD rats, standard feed, single cage, light/dark cycle:12h/12h,adaptive feed for1week,24hour fasting before stress, free of drinking water.2.Use water-immersion and restraint stress induced ulcer model in rats.3.Seventy-eight healthy male SD rats were randomly divided into4groups asfollowing: Control group(CP),Model group(MP), Ranitidine group(RP) and Xiaotan Heweidecoction group(XP).Except the CP, each of the other three groups were randomly dividedinto4groups, according to the time that the rats were released for after water-immersionand restraint stress induced, including0hourã€3hoursã€6hours and12hours. The rats in theMP were given2ml0.9%sodium chloride via oral gavage, and the RP and XP were treated with the same volume of water solution of ranitidine (50mg/kg) and XiaotanHewei decoction (2.448g/kg), respectively.4. Rats in each group were killed, and the ulcer index was observed. The apoptosiswas detected by TUNEL. The expression of glucose-regulated protein78(GRP78) andCaspase-12were both detected by immunohistochemistry and western blot.Results1. In different group that the rats were released at the same time,the gastric ulcerindex in the XP group is lower than in the MP group (P<0.05);2. In different group that the rats were released at the same time, the apoptosis indexin the XP group is lower than in the MP group (P<0.05);3. In different group that the rats were released at the same time, the expression ofGRP78in the XP group is higher than in the MP group (P<0.05), besides, theexpression of Caspase-12in the XP group is lower than in the MP group (P<0.05);ConclusionThe Xiaotan Hewei decoction can effectively reduced the ulcer index and apoptosisindex in the water-immersion and restraint stress induced rats, and the Xiaotan Heweidecoction can effectively improve the expression of protein GRP78and reduced theexpression of protein Caspase-12. The mechanism of Xiaotan Hewei decoction inpreventing the gastric mucosal cell apoptosis in water-immersion and restraint stressinduced rats may be on the base of improving the expression of protein GRP78andreducing the expression of protein Caspase-12. |
